摘 要:[目的]研究奇异海蟑螂不同提取部位对巨噬细胞RAW264.7的体外抗炎活性及对筋伤大鼠的治疗作用。[方法]以脂多糖诱导细胞株RAW264.7建立细胞炎症模型,用MTT法检测药物对RAW264.7细胞的毒性作用,Griess反应检测细胞上清液中NO的水平,进而评价奇异海蟑螂不同提取部位的体外抗炎活性。以重物自由落体组织损伤法建立筋伤大鼠模型,造模成功24 h后予以大鼠外用涂抹给药,每天1次,连续给药5 d。通过检测前列腺素E2(PGE2)、白细胞介素-1β(IL-1β)、丙二醛(MDA)和超氧化物歧化酶(SOD)等含量,观察病理切片,检测全血(高/中/低切)黏度、血浆黏度和纤维蛋白原含量等来评价奇异海蟑螂不同提取部位对筋伤模型大鼠受损肌肉组织和血液流变学的改善作用。[结果]与模型组相比,奇异海蟑螂乙醇提取物、乙酸乙酯提取物和正丁醇提取物均能显著降低RAW264.7炎症细胞释放NO含量(P<0.01),且高剂量组均能显著降低筋伤模型大鼠受损肌肉组织中PGE2、IL-1β、MDA的含量,提高受损肌肉组织的SOD活力,降低全血(高/中/低切)黏度、血浆黏度和纤维蛋白原含量(P<0.05或P<0.01),改善筋伤模型大鼠损伤肌肉的组织形态。[结论]奇异海蟑螂乙醇、乙酸乙酯和正丁醇提取物均具有显著的体外抗炎活性,同时高剂量的奇异海蟑螂不同提取部位能更为显著地增加SOD含量,降低MDA、PGE2和1L-1β的含量,降低全血(高/中/低切)黏度、血浆黏度和纤维蛋白原含量,进而达到改善模型大鼠筋伤的作用。[Objective]To study the anti-inflammatory activity of different extracts of Ligia exotica extract on macrophage RAW264.7 in vitro and the therapeutic effect on rats with tendon injury.[Methods]The cell inflammation model was established by lipopolysaccharide-induced cell line RAW264.7.The cytotoxic effect of the drug on RAW264.7 cells was detected by MTT method,and the level of NO in the cell supernatant was detected by Griess reaction,and then the anti-inflammatory activity of different extracts of Ligia exotica was evaluated in vitro.The rat model of tendon injury was established by free fall tissue injury method.After 24 hours of successful modeling,the rats were treated with external application once a day for 5 days.The contents of prostaglandin E2(PGE2),interleukin-1β(IL-1β),malondialdehyde(MDA)and superoxide dismutase(SOD)were detected,and the pathological sections were observed.The whole blood(high/medium/low shear)viscosity,plasma viscosity and fibrinogen content were detected to evaluate the improvement effect of different extracts of Ligia exotica on damaged muscle tissue and hemorheology in rats with muscle injury.[Results]Compared with the model group,the ethanol extract,ethyl acetate extract and n-butanol extract of Ligia exotica could significantly reduce the content of NO released by RAW264.7 inflammatory cells(P<0.01),and the high-dose group could significantly reduce the content of PGE2,IL-1βand MDA in the damaged muscle tissue of the rat model of muscle injury,increase the activity of SOD in the damaged muscle tissue,reduce the whole blood(high/medium/low shear)viscosity,plasma viscosity and fibrinogen content(P<0.05 or P<0.01),and improve the tissue morphology of the damaged muscle of the rat model of muscle injury.[Conclusion]The ethanol,ethyl acetate and n-butanol extracts of Ligia exotica have significant anti-inflammatory activity in vitro.At the same time,high dose of different extracts of Ligia exotica can significantly increase the content of SOD,reduce the content of MDA,PGE2 and 1L
