摘  要:目的:探讨增液承气汤加减对慢性传输型便秘治疗的作用及其作用机制。方法:健康SD大鼠60只,应用复方地芬诺酯片灌胃构建慢性传输型便秘模型大鼠,造模成功后随机分成模型组、增液承气汤加减高、中、低剂量组(剂量分别为38.88、19.44、9.72 mg/kg)、阳性药组(12.5%的伊托必利混悬液,54 mg/kg),每组10只。另取未经造模的10只SD大鼠作为正常组。正常组和模型组大鼠给予等量的超纯水,各组大鼠连续灌胃7 d。给药期间每日定期监测大鼠体质量变化、活动状态、摄食量、饮水量;给药结束后测定大鼠排便情况、肠道传输功能;ELISA法检测血清中5-HT、VIP、SP水平;Western Blotting法检测大鼠结肠组织SCF、c-kit的表达水平。结果:与模型组比较,增液承气汤加减高、中、低剂量组和阳性药组大鼠在给药期间体质量增长明显(P<0.05),摄食量增加(P<0.05),饮水量增加(P<0.05);大鼠首粒黑便排出时间明显缩短(P<0.05),血清中5-HT、VIP、SP水平明显降低(P<0.05);12 h大便数量、粪便含水量、肠管总长度、碳末推进长度、碳末推进率均明显升高(P<0.05);SCF、c-kit蛋白相对表达水平明显升高(P<0.05)。结论:增液承气汤加减可剂量依赖性地改善大鼠慢性传输型便秘,其机制可能与SCF/c-kit信号通路有关。Objective:To investigate the effect and mechanism of modified Zengye Chengqi decoction(MZCD)on the treatment of chronic transit constipation(CTC).Methods:60 SD rats were taken,50 of which were gavaged with compound diphenoxylate tablets to establish CTC models,and after the models were successfully established,they were randomly divided into model group,high,medium and low dose groups of MZCD(38.88,19.44,9.72 g/kg),and positive drug group(12.5%itopride suspension,54 mg/kg),with ten rats in each group.The rest 10 SD rats were selected as control group.The rats in the control group and the model group were given the same amount of ultrapure water,and the rats in each group were gavaged for 7 days.Furthermore,the rats were regularly monitored daily during administration for changes in body weight,activity status,and quantity of food intake and water intake.Defecation condition and intestinal transport function of the mice were observed after the administration.The levels of 5-HT,VIP,and SP in serum were detected by ELISA,and the protein expression of SCF and c-kit in colon tissue were detected by Western Blotting.Results:Compared with the model group,the rats in the MZCD groups of low,medium and high doses and the positive drug group showed significant weight gain(P<0.05),increased food intake(P<0.05),and increased water intake(P<0.05).The excretion time of the first melena in rats was significantly shortened(P<0.05),and the levels of 5-HT,VIP and SP in serum decreased significantly(P<0.05).The 12 h defecation volume,fecal water content,total intestinal length,carbon end-propulsion length,and carbon end-propulsion rate were significantly increased(P<0.05).Furthermore,the protein relative expression level of c-kit and SCF were significantly increased(P<0.05).Conclusion:MZCD can improve CTC of rats in a dose-dependent manner,and its mechanism may be related to the SCF/c-kit signaling pathway.
