摘  要:目的:确定古代经典名方乌药汤制剂适宜的提取工艺。方法:分别采用传统工艺、传统工艺水煎液蒸馏分离挥发油、二次回流提取工艺、双提法制备不同提取液,以传统工艺提取液(传统汤剂)特征图谱、指标成分含量及出膏量为参照,比较不同提取液物质基础的异同。结果:3种工艺与传统汤剂特征图谱相似度均>0.96,出膏量均差异无统计学意义(P>0.05)。与传统汤剂20个特征峰及6个指标成分相比,传统工艺水煎液蒸馏分离挥发油中13个峰绝对峰面积显著降低,2个指标成分含量显著降低(P<0.05);二次回流提取工艺中6个峰绝对峰面积显著增加,4个指标成分含量显著增加(P<0.05);双提法中2个峰绝对峰面积显著增加,1个指标成分含量显著增加(P<0.05),其余成分无显著性变化(P>0.05)。结论:双提法与传统汤剂关键质量属性基本一致,且能保留挥发性活性物质,为适宜的提取工艺。Objective:To determine the suitable extraction process of the ancient classic prescription Wuyao Decoction.Methods:Different extracts were prepared by the traditional process,the traditional process of distilling and separating volatile oil from water decoction,the secondary reflux process and the double formulation.The characteristic atlas,index component content and paste output of traditional extract(traditional decoction)were used as reference,and the similarities and differences of material basis of different extracts were compared.Results:The similarity of the characteristic atlas of the extracts from the three processes and the traditional process was greater than 0.96,and there was no statistical difference in paste output(P>0.05).Compared with the 20 characteristic peaks and 6 index components of the traditional decoction,the absolute peak area of 13 peaks and the content of 2 index components in the traditional process of distilling and separating volatile oil from water decoction were significantly reduced(P<0.05).In the secondary reflux process,the absolute peak area of 6 peaks and the contents of 4 index components increased significantly.In the double formulation,the absolute peak area of 2 peaks and the content of 1 index component increased significantly;the other components had no significant changes(P>0.05).Conclusion:The double formulation is basically consistent with the key quality attributes of traditional decoction,and can retain volatile active substances,which is a suitable extraction process.
