摘 要:本研究旨在分析建昌马Prdm9基因锌指域的遗传多样性和结构特征。实验选用建昌马(n=40),通过PCR扩增Prdm9基因锌指域并进行克隆测序。结果显示,建昌马Prdm9锌指域中存在15种C2H2型锌指,其中8个在马属动物上还没有报道;锌指间有1~4个氨基酸差异,并发生在4个固定位点上,其中3个为正向选择位点;在建昌马中检测到16种Prdm9锌指域,大多数包含9个锌指,在群体中形成A~P共14种基因型,其中AJ基因型占明显优势,A和J(ZFD1、ZFD9)为优势等位基因;Prdm9锌指域的基因杂合度、有效等位基因数、多态信息含量高,显示丰富的遗传多态性。本研究表明,建昌马Prdm9锌指和锌指域具有丰富的遗传多样性,其结构不同于马属动物中已有的报道。This study aimed to assay the genetic diversity and structure characteristics of zinc finger domain of Prdm9 gene in Jianchang horse.PCR was used to assay the tandem zinc finger sequences of Prdm9 gene of Jianchang horses(n=40),followed by gene cloning and sequencing.Fifteen kinds of C2H2 zinc fingers were discovered in the Prdm9 from 40 Jianchang horses,eight of which has not been reported yet.There existed 1 to 4 amino acid differences among the different zinc fingers,and the mutations were located at 4 fixed sites,3 of which were positive selection sites.Sixteen kinds of Prdm9 zinc finger domains were detected in Jianchang horses, most of which contained 9 zinc fingers. A total of fourteen Prdm9 genotypes (A to P)was observed in the experimental Jianchang horses, AJ was dominant genotype, and A and J alleles (refer to ZFD1 and ZFD9,respectively) were dominant alleles. The heterozygosity, number of effective allele and polymorphism information content ofPrdm9 zinc finger domains were high. The present results show that Prdm9 of Jianchang horse has rich genetic diversity, andtheir structure characteristics of zinc fingers and zinc finger domains differ with reported studies in Equids.
