摘 要:目的探讨淫羊藿苷对模拟微重力环境大鼠骨丢失的防治作用。方法取2月龄Wistar雄性大鼠30只,采用随机数字表法分为对照组、尾吊组、淫羊藿苷组,每组10只。尾吊组、淫羊藿苷组均建立尾吊模型,淫羊藿苷组给予淫羊藿苷50 mg/kg灌胃,对照组和尾吊组则给予等体积蒸馏水,每周称体质量1次。给药28 d后麻醉下腹主动脉采血处死大鼠,检测股骨骨密度并进行生物力学检测,观察并分析胫骨的HE染色结果,进行胫骨Mirco-CT检测,测定血清骨代谢生化指标。结果尾吊组和淫羊藿苷组大鼠体质量下降,但差异无统计学意义;淫羊藿苷组骨密度、骨生物力学参数较尾吊组均有所改善;淫羊藿苷组骨小梁形态、数量与厚度较尾吊组均有显著改善;淫羊藿苷组骨钙素(osteocalcin,OCN)水平较尾吊组明显增加,抗酒石酸酸性磷酸酶5b(tartrate-resistant acid phosphatase-5b,TRACP-5b)水平较尾吊组明显降低。结论淫羊藿苷对模拟微重力环境大鼠骨丢失具有良好的防治作用。Objective To investigate the effects of icariin on the prevention and treatment of bone loss in rats in simulated microgravity environment.Methods Thirty 2-month-old Wistar male rats were randomly divided into control group,tail suspension group,and icariin group,with 10 rats in each group.The tail suspension group and icariin group were established by tail suspension method.The icariin group was given icariin 50 mg/kg by gavage,while the control group and tail suspension group were given equal volume distilled water and weighed once a week.After 28 days of administration,the rats were killed by blood sampling from the abdominal aorta under anesthesia.The femoral bone mineral density was measured and the biomechanical test was carried out.The HE staining results of the tibia were observed and analyzed.The mirco-CT test of the tibia was carried out to determine the biochemical indicators of bone metabolism in the serum.Results The body mass of rats in tail suspension group and icariin group decreased,but there was no significant difference.The bone mineral density and bone biomechanical parameters of icariin group were better than those of tail suspension group.The shape,quantity,and thickness of trabecular bone in icariin group were significantly improved than those in tail suspension group.The level of OCN in icariin group was significantly higher than that in tail suspension group,and the level of TRACP-5b was significantly lower than that in tail suspension group.Conclusion Icariin has a good preventive effects on bone loss in rats under simulated microgravity environment.
