日粮精粗比对藏系绵羊精液代谢产物的影响

摘  要:试验旨在阐明日粮精粗比对高原型藏羊公羊精液代谢产物的影响。选取发育良好且体重相近(26.58±1.07)kg的高原型藏羊公羊15只,随机分为3组,每组5个重复,每个重复1只羊。各组试验羊分别饲喂精粗比为70∶30(HM组)、50∶50(MM组)和30∶70(LM组)的日粮。预试期15 d,正式试验期90 d。结果显示,随着日粮中精料比例逐渐提高,高原型藏羊平均采精量提高、精子活力以及精子密度均有不同程度提高。饲喂精粗比为70∶30的日粮,公羊平均精子活力、精子密度最好;饲喂精粗比为50∶50的日粮,公羊平均采精量最高。各组之间两两比较共筛选出1954个差异代谢物,HM组与LM组间共获得718个,其中上调离子278个,下调离子440个;MM组与LM组间共获得855个,其中上调离子446个,下调离子409个;HM组与LM组间共获得381个,其中上调离子147个,下调离子234个。KEGG分析富集的通路主要包括嘌呤代谢、戊糖磷酸途径、脂肪酸生物合成、催乳素信号通路及氨基糖与核苷酸糖代谢等。筛选出甘氨酸、葡萄糖、乙酰辅酶A、二磷酸腺苷、α-D-半乳糖作为关键性差异代谢物。HM组藏羊精液中甘氨酸含量显著低于LM组和MM组(P<0.05),其余4种关键性代谢物含量均显著高于LM组和MM组(P<0.05)。研究表明,日粮精粗比的改变能够明显地影响高原型藏羊精液的代谢及品质,以精粗比70∶30饲喂效果最好。The experiment was to elucidate the effect of dietary concentrate to roughage ratio on semen metabolites of plateau Tibetan ram.Fifteen plateau Tibetan rams with good growth and similar body weight(26.58±1.07)kg were selected and randomly divided into three groups,with five replicates in each group and one ram in each replicate.Rams in each group were fed with diets concentrate to roughage ratio of 70∶30(HM group),50∶50(MM group)and 30∶70(LM group)respectively.Pre-test period was 15 d and trial period was 90 d.The results showed that with the gradual increase of concentrate proportion in diet,the average sperm collection,sperm motility and sperm density of plateau Tibetan sheep were increased to varying degrees.The average sperm motility and sperm density of rams were the best when concentrate to roughage ratio was 70∶30.Rams fed a 50∶50 concentrate to roughage diet had the highest mean sperm intake.A total of 1954 differential metabolites were screened by two-by-two comparisons between the groups of this histological data.718 were obtained between HM group and LM group,of which 278 were up-regulated ions and 440 down-regulated ions.855 were obtained between MM group and LM group,of which 446 were up-regulated ions and 409 down-regulated ions.381 were obtained between HM group and LM group,of which 147 were up-regulated ions and 234 were down-regulated ions.The enrichment pathways of KEGG analysis mainly include purine metabolism,pentose phosphate pathway,fatty acid biosynthesis,prolactin signaling pathway,amino sugar and nucleotide sugar metabolism,etc.Glycine,glucose,acetyl-CoA,ADP and alpha-D-galactose were selected as the key differential metabolites.The content of glycine in semen of Tibetan sheep in HM group was significantly lower than that in LM group and MM group(P<0.05),and the content of other four key metabolites in semen of Tibetan sheep in HM group were significantly higher than that in LM group and MM group(P<0.05).The study indicates that the change of dietary concentrate to roughage ra
