摘 要:【目的】探讨色象中药对黑素的体外调控机制。【方法】筛选黑药(首乌、熟地、乌梅、女贞子、黒芝麻、丹参、鸡血藤)和白药(白芷、白附子、白茯苓、白蔹、白芨、白僵蚕、白术),制备各中药含药血清,体外处理人A375黑素瘤细胞16~48 h。采用四甲基偶氮唑盐(MTT)法测定黑素瘤细胞增殖情况;采用NaOH溶解法测定黑素瘤细胞黑素含量;采用免疫荧光染色法测定黑素瘤细胞干细胞因子(SCF)、c-Kit蛋白表达。【结果】(1)分别含首乌、乌梅、黑芝麻、白茯苓、白术、女贞子、白芷、白附子、白芨、白僵蚕的血清组黑素瘤细胞增殖能力较对照组升高(P<0.05或P<0.01);分别含丹参、白蔹的血清组黑素瘤细胞增殖能力较对照组降低(P<0.01)。(2)分别含熟地、黑芝麻、丹参、鸡血藤、白蔹、白术、首乌、女贞子、白芷、白附子、白芨、白僵蚕的血清组黑素瘤细胞中黑素含量较对照组增加(P<0.05或P<0.01)。(3)与对照组比较,含乌梅血清组SCF、c-Kit蛋白表达量均增加(P<0.01),分别含女贞子、白芷、白茯苓、白蔹的血清组SCF、c-Kit蛋白表达量均减少(P<0.05或P<0.01),分别含首乌、黑芝麻、丹参、白附子的血清组SCF蛋白表达量增加、c-Kit蛋白表达量减少(P<0.05或P<0.01),含白僵蚕血清组SCF蛋白表达量减少、c-Kit蛋白表达量增加(P<0.05或P<0.01),分别含熟地、鸡血藤、白术的血清组SCF蛋白表达量增加(P<0.05或P<0.01)。c-Kit蛋白表达量变化不明显(P>0.05),含白芨血清组SCF蛋白表达量变化不明显(P>0.05)。c-Kit蛋白表达量减少(P<0.01)。【结论】白药与黑药均可通过调控SCF、c-Kit表达,对黑素瘤细胞黑素代谢有双向调节作用。Objective To investigate the mechanism of in vitro regulation of melanin by color image of Chinese material medica.Methods The black color herbs(Polygoni Multiflori Radix,Rehmanniae Radix Praeparata,Mume Fructus,Ligustri Lucidi Fructus,Sesami Semen Nigrum,Salviae Miltiorrhizae Radix et Rhizoma,Spatholobi Caulis)and white color herbs(Angelicae Dahuricae Radix,Typhonii Rhizoma,Poria,Ampelopsis Radix,Bletilla Striata,Bombyx Batryticatus,Atractylodis Macrocephalae Rhizoma)were selected and the serum containing the above each drug was prepared.The proliferation of melanocytes was measured by MTT method,the melanin content of melanocytes was determined by NaOH dissolution method,and the protein expressions of stem cell factor(SCF)and c-Kit were measured by immunofluorescence staining.Results(1)The proliferation abilities of melanocytes in the groups of serum containing separately Polygoni Multiflori Radix,Mume Fructus,Sesami Semen Nigrum,Poria,Atractylodis Macrocephalae Rhizoma,Ligustri Lucidi Fructus,Angelicae Dahuricae Radix,Typhonii Rhizoma,Bletilla Striata,Bombyx Batryticatus were increased compared with the control group(P<0.05 or P<0.01);the proliferation abilities of melanocytes in the serum group containing separately Salviae Miltiorrhizae Radix et Rhizoma and Ampelopsis Radix were decreased compared with the control group(P<0.01).(2)The melanin contents of melanocytes in the groups of serum containing separately Rehmanniae Radix Praeparata,Sesami Semen Nigrum,Salviae Miltiorrhizae Radix et Rhizoma,Spatholobi Caulis,Ampelopsis Radix,Atractylodis Macrocephalae Rhizoma,Polygoni Multiflori Radix,Ligustri Lucidi Fructus,Angelicae Dahuricae Radix,Typhonii Rhizoma,Bletilla Striata,Bombyx Batryticatus were increased compared with the control group(P<0.05 or P<0.01).(3)Compared with the control group,the SCF and c-Kit protein expressions were increased in the serum group containing Mume Fructus(P<0.01),decreased in the SCF and c-Kit protein expressions in the serum groups containing separately Ligustri Lucidi Fructus,L
