摘  要:【目的】比较4种常用的SD大鼠肩周炎模型炎症因子、纤维化因子、神经内分泌因子表达,筛选有效的肩周炎造模方法。【方法】将100只SD大鼠随机分为5组,即风寒湿刺激模型组、持续机械劳损加冰敷模型组、手术损伤模型组、石膏制动模型组、空白对照组,每组20只。分别在造模完成后第1、2、4、8周取材,苏木素-伊红(HE)染色法观察肩关节软骨组织病理学形态,免疫组织化学染色法检测肩关节软骨组织炎症因子[环氧合酶1(COX-1)、肿瘤坏死因子α(TNF-α)],纤维化因子[转化生长因子β(TGF-β)],神经内分泌因子[降钙素基因相关肽(CGRP)受体成分蛋白(CRCP)、泛素羧基末端水解酶(UCHL)]表达,比较不同造模方法效果的差异。【结果】HE染色结果显示,4个模型组肩关节软骨细胞减少,关节表面软骨纤维化。免疫组织化学染色结果显示:4个模型组COX-1表达最高值均高于空白对照组最高值。风寒湿刺激模型组、手术损伤模型组TNF-α表达在造模完成后第1~2周高于空白对照组,持续机械劳损加冰敷模型组在造模完成后第1~4周均高于空白对照组,石膏制动模型组在造模完成后第1~8周均高于空白对照组。持续机械劳损加冰敷模型组、石膏制动模型组TGF-β在造模完成后第2周时最高值大于其他各组最高值。风寒湿刺激模型组、持续机械劳损加冰敷模型组、手术损伤模型组CRCP表达在造模完成后第2周最高值高于空白对照组,石膏制动模型组在第1周最高值高于空白对照组。4个模型组在造模完成后第1、2、4、8周UCHL表达均高于空白对照组。【结论】石膏制动模型在炎症因子表达稳定性方面优于其他肩周炎模型;持续机械劳损加冰敷模型、石膏制动模型在肩关节纤维化相关因子稳定表达方面优于其他模型;手术损伤模型和石膏制动模型在肩关节致痛因子表达方面优于其他模型。Objective To screen an effective method for scapulohumeral periarthritis by further comparing the expressions of inflammatory factors,fibrotic factors and neuroendocrine factors in four commonly used SD rat models of scapulohumeral periarthritis.Methods One hundred SD rats were randomly divided into five groups,namely,wind-cold dampness stimulation model group,continuous mechanical strain plus icing model group,surgical injury model group,plaster braking model group and blank control group,with 20 rats in each group.The cartilage histopathology of the shoulder joint was observed by hematoxylin-eosin(HE)staining at weeks 1,2,4 and 8 after the completion of moulding,and the inflammatory factors[cyclooxygenase 1(COX-1),tumour necrosis factorα(TNF-α)],fibrotic factor[transforming growth factor(TGF-β)],neuroendocrine factors[calcitonin gene-related peptide(CGRP)receptor component protein(CRCP),ubiquitin carboxyl-terminal hydrolase L(UCHL)]expression to compare the differences in the effects of different modelling methods.Results The results of HE staining showed that the chondrocytes of the shoulder joint were reduced and the cartilage on the joint surface was fibrotic in the four model groups.The results of immunohistochemical results staining showed as follows:The highest value of COX-1 expression in all four model groups was higher than the highest value in the blank control group.The expression of TNF-αin the wind-cold dampness stimulation model group and the surgical injury model group was higher than that in the blank control group at the weeks 1 and 2 after the completion of moulding,and that in the continuous mechanical strain plus icing model group was higher than that in the blank control group at the weeks 1-4 after the completion of moulding,and that in the plaster braking model group was higher than that in the blank control group at the weeks 1-8 after the completion of moulding.The highest value of TGF-in the continuous mechanical strain plus icing model group and the plaster braking model group was
