摘  要:目的子宫内膜异位症(endometriosis)是女性常见的疾病。最近,越来越多的证据表明miRNA在子宫内膜异位症的诊断和治疗中发挥着重要作用。然而,miR-515-5p的调控机制尚未得到充分探索。方法本研究进行了CCK-8、EdU、流式细胞术、划痕愈合和Transwell实验等探究miR-515-5p对子宫内膜细胞生物学功能的影响。此外,反转录定量qRT-PCR和Western blot分别用于评估miR-515-5p和Nrf2/Keap1通路相关蛋白的表达。构建子宫内膜异位症大鼠模型探究miR-515-5p在子宫内膜异位症病变中的作用。结果异位子宫内膜组织中miR-515-5p的表达减少,miR-515-5p的过度表达显著抑制CRL-7566细胞的增殖、迁移和侵袭。此外,miR-515-5p对CRL-7566细胞的增殖、迁移和侵袭的抑制作用是通过抑制Nrf2/Keap1通路活性所致。最后,腹腔内注射miR-515-5p mimic显著减轻了体内子宫内膜异位症病变。结论miR-515-5p通过Nrf2/Keap1通路抑制异位子宫内膜细胞增殖、迁移和侵袭。因此,miR-515-5p/Nrf2/Keap1信号通路可能是子宫内膜异位症治疗的潜在靶点。Objective Endometriosis is a common disease of women.Recently,more and more evidence has shown that miRNA has played an important role in diagnosis and treatment of endometriosis.However,the regulation mechanism of miR-515-5p has not been fully explored.Methods In this study,the effects of CCK-8,EdU,streaming cells,scratch healing,and Transwell experiments were conducted on the influence of miR-515-5p endometrial cell biological functions.In addition,the reversal volume qRT-PCR and Western blot are used to evaluate the expression of miR-515-5p and Nrf2/Keapl-channel related proteins.Build the role of uterine endometriosis rat model to explore the role of miR-515-5p in the endometriosis disease.Results The expression of miR-515-5p in the atomic endometrial tissue decreased,and the excessive expression of miR-515-5p significantly inhibited the proliferation,migration and invasion of CRL-7566 cells.In addition,the inhibitory effect of miR-515-5p on the proliferation,migration and invasion of CRL-7566 cells is caused by inhibiting the activity of Nrf2/Keapl channels.Finally,the injecting miR-515-5p mimic in the abdominal cavity significantly reduces the endometriosis disease in the body.Conclusion miR-515-5p inhibits ectopic endometrial cell proliferation,migration,and invasion through the Nrf2/Keapl pathway.Therefore,miR-515-5p/Nrf2/Keapl signaling pathway may be a potential target for the treatment of endometriosis.
