摘 要:目的 通过基于基因表达数据库(GEO)的基因表达数据,描述miR-20a-3p、miR-449b-5p与妊娠糖尿病之间的关系。方法 本研究选择了GEO数据库中的GSE182737及GSE98403两个数据集,将妊娠糖尿病组(GDM)和正常组(normal)的外周血标本分别作为实验组和对照组,分别筛选表达差异cicrRNA及miRNA,通过cicrRNA数据库筛选靶向miRNA,取交集miRNA。利用mirPATH软件对交集的差异miRNA行京都基因与基因组百科全书(KEGG)、基因本体(GO)功能分析。收集2019年1月至2020年12月在常熟市第二人民医院产科就诊的GDM的患者60例,同期正常妊娠的产妇60例,对采集的血液标本进行qPCR验证miR-20a-3p、miR-449b-5p的表达。结果 GSE182737的差异cicrRNA为10个,GSE98403的差异miRNA为73个,两个数据集表达差异miRNA交集为7个。通过miPATH软件对交集的差异miRNA行KEGG富集,结果显示在脂肪酸代谢及合成等通路上显著富集。GO富集分析显示在应激反应及细胞死亡等通路上显著富集。通过实时免疫荧光定量聚合酶链反应(qPCR)的验证,miR-20a-3p在GDM组血液样本中表达为0.688±1.024,在正常组中的表达量为3.572±1.021,miR-449b-5p在正常组血液样本中表达为11.347±1.963,在GDM组中的表达量为1.902±1.814,两组之间存在显著表达差异(P<0.05)。受试者操作特征(ROC)曲线显示miR-20a-3p、miR-449b-5p在GDM中的曲线下面积分别为0.9431及0.9398。相关性分析显示胰岛素抵抗指数与miR-20a-3p、miR-449b-5p的关系为负相关(P<0.05)。结论 根据生物信息学分析,我们发现筛选出的候选miR-20a-3p、miR-449b-5p与GDM的发生及发病机制密切相关,可以作为早期筛查GDM的生物标志物。Objective To describe the relationship between miR-20a-3p,miR-449b-5p and gestational diabetes mellitus through gene expression data based on gene expression database(GEO).Methods In this research,two datasets GSE182737 and GSE98403 in the GEO database were selected,and the peripheral blood samples of the GDM group and the normal group were used as the experimental group and the control group,respectively,and the target miRNAs of cicrRNAs were screened by the cicrRNA database.Then mirPATH software was used to analyze the functions of intersections of different mirnas in the Kyoto encyclopedia of genes and genomes(KEGG)and gene ontology(GO).A total of 60 patients with GDM and 60 normal pregnant women who were treated in the obstetrics department of Changshu No.2 People's Hospital from January 2019 to December 2020 were collected.The collected blood samples were subjected to qPCR to verify the expression of miR-20a-3p and miR-449b-5p.Results There were 10 differential cicrRNAs in GSE182737,73 differential miRNAs in GSE98403,and 7 in the intersection of expression differential miRNAs between the two datasets.The results of KEGG enrichment by miPATH software showed significant enrichment in pathways such as fatty acid metabolism and synthesis.GO enrichment analysis showed significant enrichment in pathways such as stress response and cell death.Verified by qPCR,the expression of miR-20a-3p in the blood samples of the GDM group was 0.688±1.024,the expression level in the normal group was 3.572±1.021,and the expression of miR-449b-5p in the blood samples of the normal group was 11.347±1.963,the expression level in the GDM group was 1.902±1.814,and there was a significant difference in expression between the two groups(P<0.05).The receiver operator characteristic(ROC)curve showed that the areas under the curve of miR-20a-3p and miR-449b-5p in GDM were 0.9431 and 0.9398,respectively.Correlation analysis showed that the relationship between insulin resistance index and miR-20a-3p and miR-449b-5p was negatively correla
