摘  要:目的探究一种构建产前支气管肺发育不良(BPD)动物模型的新方法,并使用肺部超声评分(LUS)对模型肺部病变的严重程度进行半定量评估。方法超声引导下穿刺胎鼠右肺并注射脂多糖(LPS)构建模型,仔鼠出生后行LUS。采集标本用于病理评分和肺泡表面活性物质相关蛋白(SP)-C及血管内皮细胞生长因子(VEGF)表达水平的检测。分析仔鼠LUS与病理评分之间的相关性。结果(1)动物模型呈现BPD样表现;(2)LUS与病理评分之间呈较强正相关性(r=0.85,P<0.005);(3)随着BPD严重程度的增加,LUS升高(P<0.05),而SP-C和VEGF的表达水平降低(均P<0.005)。结论超声引导下穿刺胎鼠肺部并注射LPS可用于构建产前BPD动物模型,LUS可用于半定量评估模型BPD的严重程度。Objective To explore a new method to construct animal model of prenatal broncho-pulmonary dysplasia(BPD),and evaluate the severity of lung lesions in the BPD model semi-quantitatively by using lung ultrasound score(LUS).Methods Ultrasound-guided puncture of the right lung of fetal rats and injection of lipopolysaccharide(LPS)was performed to construct the models,then LUS was performed on the neonate rats.The specimens were collected to detect the pathological scores and the expression levels of pulmonary surfactant-associated protein(SP)-C and vascular endothelial cell growth factor(VEGF).The correlation between LUS and pathological scores was analyzed.Results(1)The animal models demonstrated BPD-like performance.(2)There was a strong positive correlation between LUS and pathological scores(r=0.85,P0.005).(3)With the increase of BPD severity,LUS increased(P0.05),while the expression levels of SP-C and VEGF decreased(P0.005).Conclusions Ultrasound-guided puncture of the fetal rat lung and injection of LPS can be used to construct animal model of prenatal BPD,and LUS can be used to evaluate the severity of BPD in models semi-quantitatively.
