四味大发散加减方调控泪腺组织Bax/Caspase-9/Caspase-3信号通路对寒燥证干眼大鼠的作用机制

摘  要:目的:通过观察四味大发散加减方对寒燥证干眼大鼠泪液分泌(SⅠT)、泪膜破裂时间(BUT)、眼表荧光素染色(FL)的影响评估其疗效及作用机制。方法:24只大鼠随机分为空白组、模型组、中药治疗组和灸法对照组。除空白组正常喂养,其余3组造寒燥模型,中药治疗组和灸法对照组给予相应干预。在各时间点对大鼠行BUT、SⅠT及FL等检测。第28天处死大鼠,取大鼠泪腺、睑板腺组织,Western blotting检测Bax、Bcl-2、Caspase-9和Caspase-3蛋白表达,ELISA检测白介素(IL)-6水平。结果:中药治疗组和灸法对照组在治疗后泪液分泌量较治疗前显著增加(P<0.05)。大鼠眼表损伤可见恢复,舌苔由燥转润。Western blotting结果显示,泪腺组织中Bax、Caspase-9蛋白表达被明显下调,Caspase-3有下调趋势。ELISA结果显示,IL-6的水平在模型组中最低。结论:四味大发散加减方可通过抑制泪腺组织Bax/Caspase-9/Caspase-3信号通路,中止凋亡程序来增加其泪液分泌,延长泪膜破裂时间,修复角膜。Objective:To evaluate the effect and mechanism of modified Siwei Dafa Powder on the tear secretion(SIT),tear film break-up time(BUT),and fluorescein staining(FL)of dry eye rats with cold-dryness syndrome.Methods:Twentyfour rats were randomly divided into blank group,model group,traditional Chinese medicine treatment group(TCM group)and moxibustion control group(MC group).The blank group was fed normally,and the other three groups were given cold and dry models.TCM group and MC group were given corresponding intervention at the same time.The rats were tested for BUT,SIT and FL at each time point.The rats were killed on the 28th day.The lacrimal gland and meibomian gland tissues of the rats were taken.The expression of Bax,Bcl-2,Caspase-9 and Caspase-3 proteins were detected by Western blotting,and the level of IL-6 was detected by ELISA.Results:The SIT value of the TCM group and the MC group significantly increased after treatment compared to before treatment(P<0.05).The eye surface damage of rats could be recovered,and the tongue coating changed from dry to moist.Western blotting results showed,the expression of Bax and Caspase-9 proteins in rat lacrimal gland tissue was significantly downregulated,while Caspase-3 showed a downward trend.ELISA results showed that the level of IL-6 was the lowest in the model group.Conclusion:The modified Siwei Dafa Powder can increase the tear secretion,prolong the tear flm rupture time and repair the cornea by inhibiting the Bax/Caspase-9/Caspase-3 signal pathway of lacrimal gland tissue and stopping theapoptosis process.
