摘 要:目的:观察金匮肾气丸对肾阳虚大鼠空间学习记忆和Tau蛋白磷酸化的影响。方法:选用32只雄性SD大鼠,随机分为对照组、肾阳虚组、金匮肾气丸治疗组、理中丸治疗组,每组8只。除对照组外,其余采用皮下注射氢化可的松(15 mg/kg),每天1次,连续14 d,建立肾阳虚模型。造模结束后,灌胃相应药物治疗2周。结果:与对照组比较,肾阳虚组大鼠体质量下降,血浆环磷酸鸟苷(cGMP)含量增加(P<0.01),cAMP/cGMP比值减低(P<0.01)。金匮肾气丸治疗组可改善肾阳虚大鼠体质量减少。与肾阳虚组比较,金匮肾气丸治疗组和理中丸治疗组cGMP含量显著减少(P<0.05,P<0.01),理中丸治疗组cAMP/cGMP比值显著增加(P<0.01)。金匮肾气丸治疗组和理中丸治疗组较肾阳虚组找到隐藏平台的潜伏期显著缩短(P<0.01);在空间探索实验中,金匮肾气丸治疗组可显著增加穿越原平台次数(P<0.01)。肾阳虚组大鼠海马Tau蛋白苏氨酸205和丝氨酸262、396、404位点磷酸化水平较对照组显著增强,而金匮肾气丸治疗组和理中丸治疗组可显著降低Tau蛋白过度磷酸化(P<0.01)。结论:金匮肾气丸可改善肾阳虚模型大鼠肾阳虚证候,提高空间学习记忆能力。金匮肾气丸和理中丸降低Tau蛋白过度磷酸化。Objective:To observe the effects of Jingui Shenqi Pills on spatial learning and memory and Tau phosphorylation in rats with kidney-yang syndrome deficiency(KDS-Yang).Methods:Thirty-two male SD rats were randomly divided into control group,KDS-Yang group,Jingui Shenqi Pills treatment group(JS group),and Lizhong Pills treatment group(LZ group)with 8 rats in each group.A classical model of KDS-Yang in rats was induced by hydrocortisone(15 mg/kg)subcutaneous injection once a day for 14 days.After modeling,administration was performed for 2 weeks.Results:Compared with control group,changes in KDS-Yang group were significant,including decreased weight,elevated the lever of cGMP(P<0.01),and decreased the ratio of cAMP/cGMP(P<0.01).JS group could increase the weight of KDS-Yang rats.Compared with KDS-Yang group,the level of cGMP decreased in both JS and LZ group(P<0.05,P<0.01),but the ratio of cAMP/cGMP only increased in LZ group(P<0.01);The latency of finding the hidden plaform in both JS and LZ group were shorter(P<0.01);In spatial probe test,the number of times to cross the original platform JS group increased(P<0.01).Compared with control group,the phosphorylation levels of Tau at threonine 205 and serine 262,396,404 in hippocampal in KDS-Yang group enhanced(P<0.01),while the JS and LZ group could reduce the hyperphosphorylation level of Tau(P<0.01).Conclusion:Jingui Shenqi Pills could ameliorate the KDS-Yang and improve the capacity of spatial learning and memory in rats,and Jingui Shenqi Pills and Lizhong Pills could reduce the hyperphosphorylation level of Tau protein.
