摘  要:目的:探究自拟方柴黄胃溃宁对肝郁脾虚证胃溃疡大鼠肠道菌群的影响。方法:采用多因素复合模拟中西医结合乙酸法建立肝郁脾虚证胃溃疡大鼠模型。将18只SD大鼠随机分为空白组、模型组和中药组,每组6只;ELISA检测肝郁脾虚证指标;HE染色观察胃窦组织病理学改变;提取大鼠粪便肠道菌群DNA,利用16S rDNA测序观察肠道菌群变化。结果:模型组大鼠血清NE含量显著高于空白组(P<0.01),GAS、5-HT含量显著低于空白组(P<0.01);HE染色提示柴黄胃溃宁干预后可以减轻胃窦组织炎症反应,增强胃窦组织修复能力;16S rDNA测序发现,柴黄胃溃宁干预后可以显著提高肠道菌群的丰富度及多样性指数,调节菌群物种数量及其组成结构;LEf Se分析,柴黄胃溃宁可以显著提高卷曲乳杆菌属(Lactobacillus_crispatus)、支原体目(Mycoplasmatales)、坦纳菌科(Tannerellaceae)等相对丰度;降低优杆菌属(Eubacterium)、HT002、脱硫弧菌(Desulfovibrio)等相对丰度;PICRUSt2功能预测,柴黄胃溃宁对肝郁脾虚证胃溃疡大鼠肠道菌群调节功能主要富集途径是感染性疾病、细胞生长与死亡、酶家族、免疫系统疾病、折叠分类和降解、复制与修复等方面。结论:柴黄胃溃宁能够增强大鼠肝郁脾虚证胃溃疡胃窦组织修复能力,其机制可能是通过调节肠道菌群数量和组成结构并改善其富集途径。Objective:To explore the effect of Chaihuang Weikuining on the intestinal flora of rats with gastric ulcer rats with liver depression and spleen deficiency syndrome.Methods:A rat model of gastric ulcer with liver depression and spleen deficiency syndrome was established by multi-factor composite simulation of acetic acid method of integrated traditional Chinese and Western medicine.Eighteen SD rats were randomly divided into blank group,model group and traditional Chinese medicine group,with 6 rats per group.ELISA was used to detect the model indexes of liver depression and spleen deficiency syndrome;HE staining was used to observe the pathological changes of gastric antrum.The intestinal flora DNA of rats was extracted and the changes of intestinal flora were observed by 16S rDNA sequencing.Results:Serum NE levels of rats in the model group were significantly higher than those in the blank group(P<0.01),and GAS and 5-HT levels were significantly lower than those in the blank group(P<0.01);HE staining showed that Chaihuang Weikuining could reduce the inflammatory reaction of gastric antrum tissue and enhance the repair ability of gastric antrum tissue.16S rDNA sequencing revealed that the intervention of Chaihuang Weikuining gastric ulcer could significantly increase the richness and diversity index of intestinal flora,and regulate the number of species and their composition of the flora;LEfSe analysis showed that Chaihuang Weikuining gastric ulcerin could significantly increase the relative abundance of Lactobacillus_crispatus,Mycoplasmatales,and Tannerellaceae;and decrease the relative abundance of Eubacterium,HT002,Desulfovibrio.PICRUSt2 function predicted that the main enrichment pathways of Chaihuang Weikuining gastric ulcer related to the regulation of intestinal flora in rats with gastric ulcers due to liver depression and spleen deficiency were infectious diseases,cell growth and death,enzyme family,immune system disease,folding classification and degradation,replication and repair,etc..Conclusion:Chaihua
