摘 要:目的 提升天胡荽药材的质量标准。方法 参考《安徽省中药饮片炮制规范(2019年版)》对10批药材样品进行性状鉴别;对茎横切面进行显微鉴别;根据2020年版《中国药典(四部)》通则项下方法测定水分、总灰分、酸不溶性灰分、浸出物;采用薄层色谱法进行定性鉴别;采用高效液相色谱法测定槲皮素的含量,色谱柱为Agilent Zorbax Eclipse C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.4%磷酸溶液(50∶50,V/V),流速为1.0 mL/min,检测波长为360 nm,柱温为25℃,进样量为10μL。结果 天胡荽根呈细圆柱形,茎细而弯曲,叶多皱缩或破碎,气香,味微苦。茎横切面可见表皮细胞、分泌道、韧皮部、皮质部、髓部的显微特征。薄层色谱图中斑点清晰,分离度良好。10批药材样品的水分、总灰分、酸不溶性灰分、浸出物分别为9.0%,13.8%,4.2%,20.0%,槲皮素的平均含量为0.068%。结论 所建立的方法可用于天胡荽药材的质量控制,暂定药材的水分、总灰分、酸不溶性灰分分别不得过11.0%,15.0%,4.0%,浸出物及槲皮素(C_(15)H_(10)O_(7))含量分别不得少于16.0%,0.050%。Objective To improve the quality standard of Hydrocotyle sibthorpoides.Methods According to the Anhui Province Traditional Chinese Medicine Slice Processing Standards(Edition 2019),morphological identification was conducted on 10 batches of medicinal samples,microscopic identification was conducted on the transverse section of the stem.According to the general principles of the Chinese Pharmacopoeia(Edition 2020,VolumeⅣ),the water content,total ash content,acid-insoluble ash content,and extract were determined.Qualitative identification was conducted by the thin-layer chromatography(TLC)method.The content of quercetin was determined by the high-performance liquid chromatography(HPLC)method,the chromatographic column was Agilent Zorbax Eclipse C_(18)column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.4%phosphoric acid solution(50∶50,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 360 nm,the column temperature was 25℃,and the injection volume was 10μL.Results Hydrocotyle sibthorpoides hadcylindrical roots,thin and curved stems,and many wrinkled or broken leaves.It had a fragrant aroma and a slightly bitter taste.Microscopic features of the epidermis cells,secretory ducts,phloem,cortex,and pith could be seen on the stem transection.The TLC spots were clear with good separation.The water content,total ash content,acid-insoluble ash content,and extract of 10 batches of medicinal samples were 9.0%,13.8%,4.2%,and 20.0%,respectively.The average content of quercetin was 0.068%.Conclusion The established method can be used for quality control of Hydrocotyle sibthorpoides.The water content,total ash content,and acid-insoluble ash content should less than or equal to 11.0%,15.0%,and 4.0%,respectively.The content of extract and quercetin(C_(15)H_(10)O_(7))should more than or equal to 16.0%and 0.050%,respectively.
