摘 要:目的 探讨雷公藤红素联合槲皮素体外对人肺癌A549和食管癌Eca9706细胞的抗癌增效作用。方法 采用四甲基偶氮唑蓝(MTT)法测试雷公藤红素配伍槲皮素前后对人肺癌A549和食管癌Eca9706细胞增殖抑制率影响,并计算半数抑制浓度(IC50)及30%抑制浓度(IC30),通过配伍槲皮素前后雷公藤红素对两种癌细胞抑制作用的分析,从单体配伍层面评价抗癌增效作用。结果 配伍IC30槲皮素后,雷公藤红素在0.25、1.25μmol/L使人肺癌A549细胞增殖抑制率显著升高(P<0.01),而在其他更高浓度时,配伍槲皮素后抑制率无统计学差异(P>0.05);配伍IC30槲皮素后,雷公藤红素在0.25、1.25及2.50μmol/L时使人食管癌Eca9706细胞增殖抑制率显著升高(P<0.01,P<0.05),而在其他更高浓度时,配伍槲皮素后抑制率均无显著差异(P>0.05)。此外,配伍槲皮素使雷公藤红素对A549、Eca9706细胞的IC50值分别降低至2.064、2.040μmol/L、IC30值分别降低至0.337、0.408μmol/L。结论 雷公藤红素联合槲皮素对肺癌和食管癌细胞均有抗癌增效作用,并均呈现出一定量效关系。Objective To investigate the anticancer synergistic effect of tripterine combined with quercetin on human lung cancer A549 and esophageal cancer Eca9706 cells in vitro.Methods Tetramethyl azole blue(MTT)method was used to test the effect of proliferation inhibition rate of tripterine before and after the compatibility of quercetin on human lung cancer A549 and esophageal cancer Eca9706 cells,and to calculate the half inhibitory concentration(IC50)and 30%inhibitory concentration(IC30).Through the analysis of the inhibitory effect of tripterine on two kinds of cancer cells before and after the combination of quercetin,the anti-cancer synergistic effect was evaluated from the monomer compatibility level.Results Compatible with IC30 quercetin,tripterine was significantly increased the proliferation inhibition rate of human lung cancer A549 cells at 0.25 and 1.25μmol/L(P<0.01),and at other higher concentrations,there was no significant difference in inhibition rate after the compatibility of quercetin(P>0.05);after compatibility with IC30 quercetin,tripterine was significantly increased the proliferation inhibition rate of human esophageal cancer Eca9706 cells in the concentration of 0.25,1.25 and 2.50μmol/L(P<0.01,P<0.05),and at other higher concentrations,there was no significant difference in inhibition rate after the compatibility of quercetin(P>0.05).In addition,the compatibility of quercetin could reduce the IC50 value of triptolide on A549 and Eca9706 cells to 2.064 and 2.040μmol/L,and the IC30 value to 0.337 and 0.408μmol/L,respectively.Conclusions Tripterine combined with quercetin has anti-cancer synergistic effects on lung cancer and esophageal cancer cells,and both show a certain dose-effect relationship.
