摘  要:大熊猫是世界生物多样性保护的旗舰物种,秦岭大熊猫是大熊猫的重要亚种种群。间充质干细胞(MSCs)是细胞治疗领域中的理想种子细胞,是大熊猫极具保护价值的种质资源。通过组织块法和胶原酶消化法处理幼龄秦岭大熊猫骨髓、牙髓和脂肪组织后,获得了各组织来源的细胞,其形态均呈长梭形,在细胞融合度达90%时均表现出漩涡状生长状态,传至第10代时增殖能力减弱;3种来源的细胞均表达MSCs的特异性标志物CD73和CD90,不表达造血干细胞的特异性标志物CD31和CD34,其中骨髓来源细胞还表达其特异性标志物SOX-2和SCFR,牙髓来源细胞表达SCFR脂肪来源细胞表达GNL3;诱导分化结果显示,各组织来源的细胞均能分化为成脂细胞、成骨细胞和成软骨细胞。表明获得了秦岭大熊猫骨髓、牙髓和脂肪组织来源MSCs。Giant panda is the flagship species of biodiversity conservation in the world,and the Qinling giant panda is an important subspecies of giant panda population.Mesenchymal stem cells(MSCs)are ideal seed cells in the field of cell therapy and germplasm resources of giant panda with great conservation value.In this study,the bone marrow,dental pulp and adipose tissue of young Qinling giant panda were treated by tissue block method and collagenase digestion method,and cells from various tissues were obtained.The shape of cells was long spindle,and they all showed a vortice-like growth state when the cell fusion degree reached 90%,and the proliferation ability was weakened when transmitted to the 10th generation.The specific markers CD73 and CD90 of MSCs were expressed in the cells of the three sources,but the specific markers CD31 and CD34 of hematopoietic stem cells were not expressed.The specific markers SOX-2 and SCFR were also expressed in the cells of bone marrow origin,and GNL3 was expressed in the cells of dental pulp origin.The results of induction differentiation showed that the cells from all tissues could differentiate into lipoblasts,osteoblasts and chondroblasts.The results indicated that MSCs derived from bone marrow,dental pulp and adipose tissue of Qinling giant panda were obtained successfully.
