摘  要:为了制备加米霉素纳米乳,并对其理化性质进行评价。充分考虑纳米乳的药效及稳定性,筛选合适的油相、表面活性剂,并利用伪三元相图筛选最佳配方制备纳米乳。用透射电镜观察加米霉素纳米乳的形态,用激光粒度分析仪测定其粒径分布范围、多分散系数等,通过留样观察测定其稳定性。结果表明,加米霉素纳米乳最佳配方中各成分比例(质量分数)为:蔗糖脂肪酸酯32.0%,加米霉素3.0%,IPM8.0%,无水乙醇4.0%,水53.0%。制备出的加米霉素纳米乳呈规则的圆球形,平均粒径13.7 nm,外观澄清透明,稳定性试验证实其稳定性良好,动物安全性试验证明在规定用量使用前提下其安全性良好。表明加米霉素纳米乳制备工艺可操作性强,质量稳定,使用安全,有较好的应用潜力。The purpose of this stduy was to establish Gamithromycin nanoemulsion formula and evaluate its quality.Considering the effect and stability of Gamithromycin nanoemulsion,the appropriate oil phase,surfactant were selected.Then the optimum formula was designed using Pseudo-ternary phase diagram.The configuration was measured by TEM,and the particle size,polydispersity and Zeta potential were measured by LPSA.The stability was tested by high speed centrifugation and optic observation.Results showed that the optimum formula of Gamithromycin nanoemulsion was composed of 32%(w/w)sucrose fatty acid esters,3.0%(w/w)gamithromycin,8.0%(w/w)IPM,4.0%(w/w)anhydrous ethanol,53.0%(w/w)distilled water.Electron microscopy showed that the Gamithromycin nanoemulsion was composed of small spherical drops with a mean diameter of 13.7 nm(25℃).Its stability was qualified,animal safety tests showed that it is safe to use at the prescribed dosage.The Gamithromycin nanoemulsion was successfully prepared with good stability,and the method of produce was simple and easy,there was a good utilization potentiality.
