摘　　要：目的分析变应性鼻炎患者中腭、肺及鼻咽上皮克隆(palate,lung and nasal epithelium clone,PLUNC)基因表达变化,探讨PLUNC基因启动子区多态位点C-1888T是否与变应性鼻炎的易感/风险相关。方法选取2021年1月至2022年2月在广西中医药大学附属瑞康医院就诊治疗的变应性鼻炎患者225例,同时选取150例正常人为对照组,抽取外周血,比较PLUNC基因在两组人群中的表达差异;鉴定变应性鼻炎患者C-1888T多态位点3种基因型,检测不同基因型的表达频率及不同基因型中PLUNC基因的mRNA表达水平。结果变应性鼻炎患者外周血中PLUNC基因的表达较正常人群降低(P=0.000);C-1888T多态位点TT基因型及T等位基因频率显著高于对照组,且TT型外周血PLUNC基因mRNA表达水平显著低于CC型及CT型,差异有统计学意义(P=0.000)。结论变应性鼻炎患者PLUNC基因表达水平下调,C-1888T多态位点为TT型的个体更易患变应性鼻炎(OR=9.375,P=0.000)。Objective To analyze the expression changes of PLUNC(palate,lung and nasal epithelium clone)gene in patients with allergic rhinitis,and to investigate whether the polymorphic site C-1888T in the promoter region of PLUNC gene is related to the susceptibility/risk of allergic rhinitis.Methods A total of 225 patients(allergic rhinitis group)diagnosed and treated for allergic rhinitis at the Ruikang Hospital Affiliated to Guangxi University of Chinese Medicine from January 2021 to February 2022 were enrolled.Simultaneously,150 healthy individuals were selected as control group.Peripheral blood samples were collected to compare the expression differences of the PLUNC gene between the two groups.The three genotypes of the C-1888T polymorphism in allergic rhinitis patients were identified,and the expression frequencies of different genotypes and mRNA expression levels of the PLUNC gene were detected.Results The expression of the PLUNC gene in peripheral blood of allergic rhinitis patients was significantly lower than that in the normal control group(P=0.000).The TT genotype and T allele frequency of the C-1888T polymorphism in the allergic rhinitis group were significantly higher than those in control group.Additionally,the mRNA expression level of the PLUNC gene in peripheral blood of individuals with the TT genotype was significantly lower than that in those with the CC and CT genotypes,with statistical significance(P=0.000).Conclusion The expression level of the PLUNC gene is downregulated in patients with allergic rhinitis.Individuals with the TT genotype of the C-1888T polymorphism are more susceptible to allergic rhinitis(OR=9.375,P=0.000)
