摘 要:目的:观察基于“壮火食气”的葛根复方对糖尿病心肌病(diabetic cardiomyopathy,DCM)大鼠心脏钙转运蛋白的影响,并讨论其干预作用。方法:采用高脂高糖饮食联合链脲佐菌素(Streptozotocin,STZ)腹腔注射进行DCM大鼠的制备,将成模大鼠随机分为模型组6.5g/(kg·d)、达美康组20mg/(kg·d)、葛根复方组8.65g/(kg·d),并选取8只空白大鼠为正常组。按上述剂量灌胃结束后采用蛋白印迹法(Western blot)检测DCM大鼠心肌组织中肌浆网钙泵ATP酶(SERCA2a)及钠钙交换体(NCX1)的表达水平、Masson染色观察DCM大鼠心肌纤维化程度,实验期间观察记录大鼠一般情况,并于给药后每日上午九点检测各组大鼠随机血糖(RBG)水平。结果:1:药物干预后,各给药组血糖均显著下降(P<0.05),其中达美康组降糖效果优于葛根复方组。2.与正常组对比,模型组SERCA2a、NCX1蛋白表达水平显著下降(P<0.01);与模型组对比,葛根复方组SERCA2a、NCX1蛋白表达水平显著上升(P<0.01)。3.与正常组对比,模型组心肌细胞排列紊乱、心肌细胞间可见胶原纤维堆积明显增加(P<0.05);与模型组对比,葛根复方组心肌细胞排列趋于整齐,胶原纤维堆积明显减少(P<0.05)。结论:基于“壮火食气”的葛根复方可以提高DCM大鼠心肌组织中SERCA2a、NCX1的蛋白表达水平、降低血糖、改善心肌纤维化,为葛根复方治疗DCM提供了新的实验依据。Objective:To observe the effect of Gegen compound based on“hyperactive fire consuming qi”on cardiac calcium transporter in diabetic cardiomyopathy(DCM)rats and discuss its intervention effect.Methods:DCM rats were prepared by intraperitoneal injection of high fat and high sugar diet combined with streptozotocin(STZ).The model rats were randomly divided into model group(6.5g/kg(d),damekang group(20mg/kg·d)and pueraria compound group(8.65g/kg·d).Eight blank rats were selected as the normal group.After intragastric administration,the expression levels of sarcoplasmic reticulum calcium pump ATPase(SERCA2a)and sodium calcium exchanger(NCX1)in myocardium of DCM rats were detected by Western blot,and the degree of myocardial fibrosis in DCM rats was observed by Masson staining.The fasting blood glucose(FBG)of rats in each group was measured at nine o'clock after administration.Results:1:After the intervention,the blood glucose of each drug group decreased significantly(P<0.05).2.Compared with the normal group,the expression level of SERCA2a and NCX1protein in the model group decreased significantly(P<0.01);Compared with the model group,the expression levels of SERCA2a and NCX1protein in Gegen compound group increased significantly(P<0.01).3.Compared with the normal group,the arrangement of myocardial cells in the model group was disordered,and the accumulation of collagen fibers between myocardial cells was significantly increased(P<0.05);Compared with the model group,the arrangement of myocardial cells in the Gegen compound group tended to be neat,and the accumulation of collagen fibers was significantly reduced(P<0.05).Conclusion:Gegen compound based on“hyperactive fire consuming qi”can increase the protein expression level of SERCA2a and NCX1in myocardial tissue of DCM rats,reduce blood sugar and improve myocardial fibrosis,which provides a new experimental basis for Gegen compound to treat DCM.
