摘 要:目的评价柔肝方对自发性高血压大鼠(SHR)降压效果及其影响大鼠血管内皮功能的作用机制和对心肌细胞超微结构的影响。方法取Wistar-Kyoto(WKY)大鼠10只作为正常组,取SHR大鼠50只分为5组,分别为模型组、厄贝沙坦组及柔肝方高、中、低剂量组。连续给药8周后测量各组大鼠血压,酶联免疫分析(ELISA)法测定血清中内皮素(ET)、一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)、细胞间黏附分子1(ICAM-1)、血管内皮细胞黏附分子1(VCAM-1)含量,实时荧光定量PCR(RT-qPCR)法检测血管内皮组织ICAM-1、VCAM-1 mRNA表达水平,透射电镜下观察各组心肌细胞超微结构的形态改变。结果柔肝方能明显降低SHR大鼠的收缩压与舒张压(P<0.01),明显降低血清中ET、MDA、ICAM-1、VCAM-1含量(P<0.05,P<0.01),明显升高NO、SOD含量(P<0.05,P<0.01),明显降低血管内皮组织ICAM-1、VCAM-1的mRNA表达(P<0.01),同时能减轻心肌超微结构的损伤。结论柔肝方对SHR大鼠具有明显的降血压作用,其作用机制与改善血管内皮功能有关,同时柔肝方可在一定程度修复受损的心肌细胞。Objective To evaluate the effect of Rougan Decoction on blood pressure in spontaneous hypertensive rats(SHR),and to explore the mechanism of its effect on vascular endothelial function and the ultrastructure of myocardial cells.Methods Ten Wistar-Kyoto(WKY)rats were taken as the normal group,while 50 SHR rats were randomly divided into five groups including the model group,the Irbesartan group,and three different dosage(low-,middle-and high-dosage)groups of Rougan Decoction.Each group was administered by continuous gavage for 8 weeks.The blood pressure was measured after 8 weeks of administration.The ELISA kit was used to detect the contents of endothelin(ET),nitric oxide(NO),superoxide dismutase(SOD),malondialdehyde(MDA),intercellular adhesion molecule-1(ICAM-1)and vascular endothelial cell adhesion molecule-1(VCAM-1)in the serum of rats.The mRNA expressions of ICAM-1 and VCAM-1 in vascular endothelial tissue were detected by RT-qPCR.The morphological change of myocardial ultrastructure was observed by transmission electron microscope.Results Rougan Decoction significantly reduced the systolic and diastolic blood pressures of SHR rats(P<0.01).Moreover,it decreased the contents of ET,MDA,ICAM-1 and VCAM-1(P<0.05,P<0.01),increased the contents of NO and SOD(P<0.05,P<0.01),significantly reduced the mRNA expressions of ICAM-1 and VCAM-1 in vascular endothelial tissue(P<0.01),and reduced the damage of myocardial ultrastructure.Conclusion Rougan Decoction can significantly reduce blood pressure in SHR rats.The mechanism was related to the improvement of vascular endothelial function.In addition,it can also alleviate the damage of myocardial ultrastructure.
