摘  要:目的分析加味小青龙汤合达原饮治疗新型冠状病毒感染(COVID-19)的临床效果,探讨其活性成分、潜在靶点和作用机制。方法比较常规治疗和加味小青龙汤合达原饮治疗COVID-19的中医证候疗效、炎症指标及不良反应,评价加味小青龙汤合达原饮治疗COVID-19的临床效果。应用TCMSP数据库收集加味小青龙汤合达原饮中活性成分,使用SwissTargetPrediction工具预测其潜在靶点;利用Pubchem,Immport等数据库获取COVID-19相关的疾病靶点,取药物-疾病交集靶点;对交集靶点进行蛋白相互作用(PPI)分析与网络构建;对交集靶点进行GO和KEGG分析;利用Autodock vina软件对其重要成分及关键靶点进行分子对接。结果经加味小青龙汤合达原饮治疗后,COVID-19患者的临床症状和炎症指标改善均优于对照组。加味小青龙汤合达原饮治疗COVID-19的相关靶点共34个,其中核心靶点有EGFR、RELA、RELA、ICAM-1、MMP9等。主要通路涉及PI3K-Akt信号通路、Ras信号通路、MAPK信号通路等。结论加味小青龙汤合达原饮可通过多成分-多靶点治疗新型冠状病毒引起的“寒湿疫”。Objective To analyze the clinical effect of Modified Xiaoqinglong Decoction and Dayuan Decoction in the treatment of COVID-19,and to explore the effective components,potential targets,and mechanism of action associated with these decoctions.Methods The study compared the efficacy of traditional Chinese medicine(TCM)syndromes,inflammatory indexs,and adverse reactions between routine treatment and Modified Xiaoqinglong Decoction and Dayuan Decoction in the treatment of COVID-19.The clinical effect of Modified Xiaoqinglong Decoction plus Dayuan Decoction in the treatment of COVID-19 was also evaluated.Active ingredients in Modified Xiaoqinglong Decoction and Dayuan Decoction were collected from TCMSP databases,then the SwissTargetPrediction tool was used to predict the potential targets of these ingredients.After the targets of COVID-19 were obtained from databases such as Pubchem,Immport,the intersection of drug targets and disease targets was conducted.Overlapping targets were identified using a protein-protein interaction network analysis(PPI),and a network was constructed.Gene Ontology(GO)function enrichment analysis and KEGG pathway enrichment analysis were used to analyze the intersecting targets.Molecular docking studies between key components and targets were performed by Autodock Vina software.Results The Modified Xiaoqinglong Decoction and Dayuan Decoction treatments were found to be more effective in improving clinical symptoms and inflammatory indicators of patients than those in the control group.The study identified 34 targets related to the treatment of COVID-19 by Modified Xiaoqinglong Decoction and Dayuan Decoction.The core targets are EGFR,RELA,ICAM-1,MMP9,etc.The main pathway involved PI3K-Akt signaling pathway,Ras signaling pathway,MAPK signal pathway etc.Conclusion The results of this study indicate that Modified Xiaoqinglong Decoction and Dayuan Decoction exhibit significant therapeutic effects on“cold-damp epidemic disease”caused by the novel coronavirus through a multi-component-multi-tar
