摘 要:本研究旨在探讨山茱萸中的活性成分马钱子苷是否对超负荷运动引起的心肌细胞焦亡具有保护作用。本研究采用递增负荷游泳法建立了超负荷运动诱导的心肌损伤大鼠模型,并将大鼠分为多个实验组,分别灌胃不同剂量的马钱子苷。通过心功能参数的检测、血清心肌损伤指标的测定和心肌组织氧化应激指标的测量,研究发现马钱子苷能够显著改善超负荷运动诱导的心肌损伤大鼠的心功能,减轻心肌损伤,并抑制心肌组织的氧化应激反应。进一步的组织学染色和RT-qPCR分析结果显示,马钱子苷可以剂量依赖性地抑制超负荷运动诱导的心肌细胞焦亡,降低TUNEL阳性率和Bax转录水平,并升高Bcl-2转录水平。通过Western blot实验,本研究证实马钱子苷可抑制超负荷运动诱导的心肌损伤大鼠心肌组织中NLRP3炎症体相关蛋白(NLRP3,cleaved caspase-1,ASC,IL-1β和IL-18)的表达水平。本研究结果初步表明,山茱萸中的活性成分马钱子苷对超负荷运动引起的心肌细胞焦亡具有显著的保护作用,可能通过抑制细胞焦亡和减轻氧化应激反应来改善超负荷运动诱导的心肌损伤。This study aimed to investigate whether the active component loganin from Cornus officinalis has a protective effect on cardiomyocyte pyroptosis induced by overloaded exercise.An overloaded exercise-induced cardiomyopathy rat model was established using the incremental load swimming method,and the rats were divided into multiple experimental groups with varying doses of loganin administered orally.Through the evaluation of cardiac function parameters,measurement of serum myocardial injury markers,and assessment of myocardial oxidative stress indicators,the study found that loganin significantly improved cardiac function,reduced myocardial injury,and inhibited oxidative stress in overloaded exercise-induced cardiomyopathy rats.Further histological staining and RT-qPCR analysis demonstrated that loganin dose-dependently suppressed cardiomyocyte pyroptosis,as evidenced by decreased TUNEL-positive cells and Bax mRNA levels,and increased Bcl-2 mRNA levels.Western blot experiments confirmed that loganin suppressed the expression of NLRP3 inflammasome-related proteins(NLRP3,cleaved caspase-1,ASC,IL-1β,and IL-18)in the heart tissues of overloaded exercise-induced cardiomyopathy rats.The preliminary findings of this study suggest that loganin,an active component of Cornus officinalis,exerts a significant protective effect against cardiomyocyte pyroptosis induced by overloaded exercise,possibly through the inhibition of pyroptosis and attenuation of oxidative stress to improve overloaded exercise-induced cardiomyopathy.
