复方功应草的急性和亚慢性毒性试验

摘  要:本试验旨在通过小鼠急性毒性试验和大鼠30 d亚慢性毒性试验对复方功应草进行安全性评价。在急性毒性试验中,取48只ICR小鼠进行预试验和最大给药剂量试验,观察小鼠表现。在亚慢性毒性试验中,将80只SD大鼠随机分为复方功应草高剂量组[17.4 g/(kg·bw)]、中剂量组[8.5 g/(kg·bw)]、低剂量组[1.7 g/(kg·bw)]和空白对照组进行灌胃给药,每天1次,连续30 d,每天观察各组大鼠的一般行为表现,记录每周平均体重、采食量和饮水量;试验结束时每组随机选择10只大鼠(雌雄各5只)采血,检测血液学和血液生化指标;将大鼠处死后采集主要脏器称重,计算脏器系数并进行组织病理学检查。结果显示,在急性毒性试验中,小鼠1 d内连续3次经口染毒累积剂量达到30 g/(kg·bw)时,无明显中毒反应,证明复方功应草实际无毒。在亚慢性毒性试验中,复方功应草高、中、低剂量组大鼠的一般行为表现均正常,无中毒和死亡情况;每周平均体重、采食量和饮水量均与空白对照组无显著差异(P>0.05);血液学、血液生化和脏器系数均在正常范围内,与空白对照组无显著差异(P>0.05);组织病理学检查无明显病理变化,对主要器官无不良影响;即复方功应草按照17.4 g/(kg·bw)(相当于靶动物猪推荐给药剂量的51.2倍)剂量连续服用30 d对大鼠安全性较高。本试验结果表明,复方功应草有较高的临床用药安全性,无毒性作用,该结果为复方功应草的临床用药提供了数据支持。The objective of this study was to assess the safety of Compound Gongying Cao through acute toxicity tests in mice and subchronic toxicity tests in rats.In the acute toxicity test,48 ICR mice were used for preliminary tests and maximum dose tests,and their behavior was observed.In the subchronic toxicity test,80 SD rats were randomly divided into Compound Gongying Cao high-dose group[17.4 g/(kg·bw)],medium-dose group[8.5 g/(kg·bw)],low-dose group[1.7 g/(kg·bw)],and blank control group,with oral gavage administration once a day for 30 days.General behavior,weekly average body weight,food consumption,and water intake were observed in each group.At the end of the experiment,10 rats(5 males and 5 females)from each group were randomly selected for blood collection to analyze hematological and blood biochemical parameters.The major organs were collected,weighed,and the organ coefficients were calculated.Histopathological examination of the tissues was also performed.The results showed that in the acute toxicity test,there were no significant toxic reactions observed in mice when the cumulative dose reached 30 g/(kg·bw)within 1 day,demonstrating that Compound Gongying Cao was non-toxic in practice.In the subchronic toxicity test,rats in the high-,medium-,and low-dose groups exhibited normal general behavior with no signs of toxicity or mortality.There were no significant differences in average weekly body weight,food consumption,and water intake compared to the blank control group(P>0.05).Hematological,blood biochemical parameters,and organ coefficients were within normal ranges and showed no significant differences compared to the blank control group(P>0.05).Histopathological examination revealed no significant pathological changes,and there were no adverse effects on major organs.Thus,Compound Gongying Cao,when administered continuously at a dose of 17.4 g/(kg·bw)(equivalent to 51.2 times the recommended therapeutic dose for pigs)for 30 days,demonstrated a high level of safety in rats.These results indicate that
