摘  要:睾丸未成熟支持细胞的增殖活性对种公畜繁殖性能有重要影响,睾丸基因组DNA甲基化(5mC)不同发育阶段特异性引起的基因差异表达可以调控支持细胞的增殖活性。在课题组前期研究中,SMAD6被鉴定出是睾丸发育过程中受3′UTR甲基化水平调控的枢纽基因之一,但其生物学功能尚未被发掘。本研究旨在探究SMAD6对猪未成熟支持细胞增殖和凋亡的作用。在体外培养的猪未成熟支持细胞中过表达SMAD6或干扰SMAD6表达后,检测细胞周期和增殖相关基因的相对表达量、细胞增殖情况、细胞ATP水平、细胞凋亡率及凋亡相关蛋白的表达情况。结果显示:过表达SMAD6后细胞周期及增殖相关基因的相对表达量上升(P<0.05或P<0.01)、细胞增殖增加(P<0.05或P<0.01)、细胞ATP水平升高(P<0.01)、细胞的凋亡率下降(P<0.05)、促凋亡相关蛋白BAX和Caspase3的表达降低(P<0.05);干扰SMAD6表达结果则与之相反。研究结果提示,SMAD6促进猪未成熟支持细胞的增殖且抑制其凋亡。The proliferative activity of immature Sertoli cells plays an important role in the reproductive performance of male breeding animals.Differential expression of genes induced by DNA methylation(5-methylcytosine,5mC)of the testis at different developmental stages regulates the proliferative activity of the Sertoli cells.In our previous research,SMAD6(SMAD family member 6)was identified as one of.the hub genes regulated by the methylation level of 3'UTR(3'-untranslated regions)during testicular development,but its biological function has not been explored.This study aimed to investigate the effects of SMAD6 on the proliferation and apoptosis of immature porcine Sertoli cells.The relative expression of the cycle and proliferation-related genes,cell proliferation,ATP level in the cells,the apoptosis rate and expression of apoptosis-related proteins were detected after over-expressing or interfering with SMAD6 expression in the immature porcine Sertoli cells cultured in vitro.The results showed that the over-expression of SMAD6 increased the expression level of the cycle and proliferation-related genes(P<0.05 or P<0.01),the cell proliferation rate(P<0.05 or P<0.01),and the ATP level(P<0.01),but decreased the apoptosis rate(P<0.05)and the expressions of pro-apoptotic related proteins BAX and Caspase3(P<0.05).However,the SMAD6 interference showed the opposite effects.In summary,SMAD6 promoted the proliferation of immature porcine Sertoli cells and inhibited their apoptosis.
