摘  要:为筛选体外抗氧化能力最强的丹参多糖组分,并研究其对小鼠抗肝肾损伤与免疫保护作用及机制,本研究通过分光光度法测定1,1-二苯基-2-三硝基苯肼自由基(DPPH·)、超氧阴离子自由基(O_(2)^(-)·)、羟基自由基(·OH)清除能力及总还原力等评价丹参多糖不同分子量组分的体外抗氧化能力。结果显示相同的条件下,丹参多糖低分子量组分具有更强的体外抗氧化能力;将60只ICR小鼠分为对照组、模型组及丹参多糖低分子量组分低、中、高剂量给药组(75 mg/kg,150 mg/kg,300 mg/kg)共5组。除对照组外,其余各组小鼠腹腔注射环磷酰胺(CTX)制备肝肾损伤及免疫抑制小鼠,给药组每日灌胃给药1次,共给药30 d。给药结束后称量小鼠体质量并剖杀各组小鼠,取肝脏、肾脏、脾脏及胸腺等脏器称重并计算脏器指数。利用血液生化仪测定小鼠肝肾功能主要生化指标(ALT、AST、TP、ALB、GLOB、T-BIL、UREA、CREA),采用分光光度法检测各组小鼠肝组织中抗氧化指标(SOD、MDA、CAT、GSH-Px)、采用流式细胞术检测小鼠血液中T细胞分型。结果显示,与对照组比,高剂量给药组小鼠能够抵抗CTX所致的体质量下降(P<0.05)。与模型组比,高剂量给药组小鼠肝脏和脾脏指数、血清AST及ALT、肝组织MDA含量极显著降低(P<0.01);血清CREA、肝组织SOD及GSH-Px活性、外周血CD3^(+)CD4^(+)T细胞亚群比例及CD4^(+)/CD8^(+)T细胞的比值极显著升高(P<0.01);胸腺指数、血清中TP、UREA及ALB含量、肝组织CAT活性显著升高(P<0.05);中剂量给药组小鼠肝脏指数及肝组织MDA含量极显著降低(P<0.01),ALT含量显著降低(P<0.05),血清TP含量显著升高(P<0.05);低剂量给药组小鼠肝脏指数显著降低(P<0.05)。以上结果表明丹参多糖低分子量组分(<10 ku)具有较高的体内外抗氧化能力,从而对CTX所致肝肾损伤及免疫抑制模型小鼠具有抗肝肾损伤及免疫保护作用。本研究为丹参水�This study aimed to screen out the polysaccharide with the highest antioxidant capacity from Salvia miltiorrhiza in vitro,and explore its effects and mechanism on liver damage,kidney damage and immunosuppression.Antioxidant capacity in vitro was evaluated by determining scavenging activity of DPPH·,O_(2)^(-)·,·OH and total reducing power of S.miltiorrhiza polysaccharides with different molecular weight.The results showed that the low-molecular-weight polysaccharides from S.miltiorrhiza exhibited the highest antioxidant activity in vitro among these components under the same conditions.Sixty ICR mice were randomly divided into five groups(n=12 in each group).The normal control group,Cyclophosphamide(CTX)model group,low-molecular-weight polysaccharides groups(75,150,300 mg/kg).Mice were injected intraperitoneally with CTX to establish liver damage,kidney damage and immunosuppression model except the normal control group.Each dose group was intragastric administration once a day for 30 days.The body weight of every mouse was measured before being killed,and their liver,kidney,spleen and thymus were weighed and the corresponding indexes were measured.The main biochemical indexes(ALT,AST,TP,ALB,GLOB,T-BIL,UREA,CREA)of liver and kidney function were measured by blood biochemical instrument,the antioxidant indexes in liver tissue(SOD,MDA,CAT,GSH-Px)were tested by spectrophotometric method,and T-lymphocyte subsets in peripheral blood of mice were measured by flow cytometry.The results showed that compared with the control group,the high dose group showed significant improvements in the loss of weight in CTX model group(P<0.05).Compared with the model group,the high dose group showed significantly decrease for the amounts of liver index,spleen index,AST and ALT in serum,MDA in liver(P<0.01);and significant improvements for the activities of CREA,SOD and GSH-Px in the serum,the amount of CD3^(+)CD4^(+)in peripheral blood,and the ratio of CD4^(+)/CD8^(+)(P<0.01);and significant improvements for the percentage of thymus i
