摘  要:以辣椒疫霉菌为试材,采用基因沉默及过表达方法,研究了磷脂酶D PcPLD8基因的功能,揭示该基因对辣椒疫霉菌生长发育及致病力的影响,以期为深入揭示磷脂酶D在辣椒疫霉菌致病中的作用机理提供参考依据。结果表明:与野生型菌株WT和空载体转化子CK相比,PcPLD8基因沉默转化子S1和S5菌丝稀疏,侧枝多且短小,菌落直径小,生长速率慢,对本氏烟的致病力明显下降;PcPLD8基因过表达转化子OE2在菌丝形态、生长速率及对本氏烟的致病力无显著影响。Phytophthora capsici was used as test material,the function of phospholipase D PcPLD8 gene was studied by gene silencing and gene overexpression,the effects of PcPLD8 gene on the growth and pathogenicity in P.capsici were revealed,in order to provide reference for further revealing the mechanism of phospholipase D in the pathogenesis of P.capsici.The results showed that the silenced transformants S1 and S5 of PCPLD8 gene had sparse hyphae,many and short lateral branches,small colony diameter and slow growth rate,the pathogenicity to Nicotiana benthamiana decreased obviously,compared with the wild-type strain WT and the empty vector transformant CK.And PCPLD8 over-expressed transformant OE2showed little change in hypha morphology,growth rate and pathogenicity to N.benthamiana.
