摘 要:以叶艺兰(Cymbidium tracyanum var.‘huanghua’×C.iridioides D.Don.‘Yeyi’)为试材,以课题组前期获得的叶艺兰上调表达CcMYB24基因的EST序列为基础,采用PCR技术扩增全长,并进行生物信息学分析和亚细胞定位,研究了CcMYB24基因功能,以期为植物MYB24的结构与功能利用提供参考依据。结果表明:CcMYB24基因全长908 bp,编码301个氨基酸,生物信息学分析预测CcMYB24蛋白是偏碱性、不稳定、亲水、非跨膜的非分泌蛋白;CcMYB24氨基酸序列具有1个特定匹配的保守MYB蛋白DNA结合域,且含有2个R结构,属于R2R3-MYBs家族;亚细胞定位结果显示CcMYB24蛋白定位在细胞核上。系统进化树表明,其与建兰(Cymbidium ensifolium)亲缘关系最近,与建兰CeRAX2-like同源性最高,为95.99%。因CcMYB24基因属于R2R3-MYBs家族,推测CcMYB24转录因子可能发挥着与R2R3-MYB转录因子相似的作用。Cymbidium with variegated leaf(Cymbidium tracyanum var.‘huanghua' × C.iridioides D.Don.‘Yeyi') was used as the experimental material,based on the EST sequence of up-regulated expression of CcMYB24 gene obtained by our research group.The full length of CcMYB24 gene was amplified by PCR,and the function of CcMYB24 gene was studied by bioinformatics analysis and subcellular localization,in order to provide reference for the utillization of the structure and fundion of plant MYB24.The results showed that CcMYB24 gene was 908 bp and encoded 301 amino acids.Bioinformatics analysis predicted that CcMYB24 protein was alkaline,unstable,hydrophilic and non-transmembrane non-secretory protein.The CcMYB24 amino acid sequence had a specific matched conserved MYB protein DNA binding domain and contains two R structures,which belonged to the R2R3-MYBs family.The results of subcellular localization showed that CcMYB24 protein was located on the nucleus.Phylogenetic tree showed that it was closely related to Cymbidium ensifolium and had the highest homology with CeRAX2-like of Cymbidium ensifolium(95.99%).Since CcMYB24 belonged to the R2R3-MYBs family,it was speculated that CcMYB24 transcription factor might play a similar role to R2R3-MYB transcription factor.
