摘 要:通过不同质量浓度细粒棘球蚴囊液(Echinococcus granulosus cyst fluid,Eg CF)体外刺激小鼠脾淋巴细胞,观察其对Th9细胞分化的影响。以100、200、400和800μg/m L Eg CF体外与小鼠脾淋巴细胞共培养后收集细胞,通过CCK8检测细胞增殖能力,流式细胞术检测各组CD3^(+)T、CD4^(+)T、CD4^(+)IL-9^(+)T细胞占比和凋亡率。CCK8检测结果显示,随着Eg CF质量浓度的增加,脾淋巴细胞增殖活性升高且成剂量依赖性(P<0.001)。细胞凋亡检测结果表明,随着Eg CF质量浓度的增加,小鼠脾淋巴细胞凋亡率增加(P<0.05)。流式细胞术检测结果显示,与对照组相比,不同质量浓度Eg CF刺激促进脾淋巴细胞中CD3^(+)T细胞和CD4^(+)IL-9^(+)T细胞的分化,抑制CD4^(+)T细胞的分化,且均成剂量依赖性(P<0.05)。结果表明,不同质量浓度Eg CF刺激通过上调Th9细胞相关细胞因子IL-9,促进细胞增殖和凋亡,调控宿主的免疫细胞,对阐明细粒棘球蚴调节宿主免疫应答提供了理论依据,可为细粒棘球蚴病免疫预防提供新的研究思路。To investigate the effect of various mass concentrations of Echinococcus granulosus cystic fluid(EgCF)on the differentiation of Th9 cells in mouse splenic lymphocytes,mouse spleen lymphocytes were cultured in vitro at various EgCF mass concentrations(100μg/mL,200μg/mL,400μg/mL and 800μg/mL).The CCK8 test was used for cellular proliferation detection.Flow cytometry was used to de-tect the proportion of CD3^(+)T,CD4^(+)T,CD4^(+)IL-9^(+)T cells and apoptosis rate in each group.The CCK8 trial showed that splenic lymphocyte proliferation activity increased with increased EgCF mass concentration as a function of dose(P<0.001).Results of detection of apoptosis showed that as the mass concentration of EgCF increased,the rate of apoptosis of spleen lymphocytes in mice increased(P<0.05).Flow cytometry showed that different mass concentrations of EgCF promoted the differentiation of CD3^(+)T cells and CD4^(+)IL-9^(+)T cells and inhibited the differentiation of CD4^(+)T cells in a dose-dependent manner compared with the control group(P<0.05).In conclusion,the stimulation of EgCF at different mass concentrations promoted cell proliferation and apoptosis through upregulation of Th9 cell-related cytokine IL-9,and regulated host immune cells,which provided theoretical significance to elucidate the regulation of host immune response by E.granulosus and could provide new research ideas for immunoprophylaxis of cystic echinococcosis.
