摘 要:基于网络药理学和分子生物学探讨白花蛇舌草对人胶质瘤U251细胞生长及细胞周期分布的影响及机制.应用CCK-8试剂盒、划痕实验和流式细胞术检测白花蛇舌草抗脑胶质瘤的作用;筛选白花蛇舌草抗肿瘤活性成分,应用网络药理学方法构建白花蛇舌草活性成分-作用靶点-疾病的相互作用网络并应用分子生物学手段解析其作用机制.研究发现,白花蛇舌草可抑制U251细胞的增殖和迁移,诱导该细胞的凋亡,阻止该细胞的周期进展从而发挥其抗脑胶质瘤作用.其作用机制可能是多种抗肿瘤活性成分通过多个信号通路影响细胞功能,其中熊果酸可抑制U251细胞的增殖并诱导细胞凋亡,伴有Bax/Bcl-2表达水平增加.为进一步研究白花蛇舌草抑制胶质瘤的药效物质基础提供了理论依据.To analyze the effects of Hedyotis diffiusa Willd(HDW)on the growth and cell cycle distribution of human glioma U251 cells based on the network pharmacology and molecular biology.The anti-glioma effects of HDW were carried out by CCK-8,scratch test and flow cytometry.The active ingredients of HDW were screened out and the active ingredients-targets-disease interaction network were set up.The biological functions and pathways involved in the targets were analyzed by molecular biology methods.Our study found that HDW extract could inhibit the proliferation and migration of U251 cells,promote the apoptosis of U251 cells,and prevent the progression of U251 cell cycle.Its mechanism may be that a variety of anti-tumor active ingredients affect cell function through multiple signal pathways.Therein ursolic acid as an main anti-tumor active ingredient of HDW could inhibit the proliferation and promote the apoptosis of U251 cells,accompanied by increased Bax/Bcl-2 protein levels.This experiment provides a certain theoretical basis for further study of the material basis of anti-glioma effect of HDW.
