摘 要:目的比较5种细菌在硫乙醇酸盐流体培养基的薄膜过滤器与试管中生长是否存在差异性。方法采用金黄色葡萄球菌、铜绿假单胞菌、枯草芽孢杆菌、生孢梭菌、洋葱伯克霍尔德菌制备1~10 cfu/mL和10~100 cfu/mL两种浓度的菌悬液,分别接种至装量为100 mL和50 mL硫乙醇酸盐流培养基的一次性薄膜过滤器,以装量为15 mL硫乙醇酸盐流体培养基的试管作对照,观察三者中细菌的生长情况。结果上述两种浓度的5种细菌悬液在装量为50 mL一次性薄膜过滤和15 mL的试管中均生长良好。在装量为100 mL一次性薄膜过滤器,菌悬液浓度为10~100 cfu/mL各菌株均生长良好,而菌悬液浓度为1~10 cfu/mL的生长检查中有2批次铜绿假单胞菌不生长,3批次枯草芽孢杆菌不生长。结论在考察硫乙醇酸盐流体培养基生长检查的工作中,建议同时选取装量为100 mL一次性薄膜过滤器,加入菌株数量为1~10 cfu/mL考察其生长情况,以降低无菌检查中假阴性出现的概率。Objective To compare the difference of growth of five bacteria in disposable membrane filter and test tube of thioglycollate fluid medium.Methods Bacterial suspensions of staphylococcus aureus,pseudomonas aeruginosa,bacillus subtilis,clostridium sporogenes,and burkholderia cepacia were prepared with concentrations of 1-10 cfu/mL and 10-100 cfu/mL,and then inoculated into disposable membrane filters containing 100 mL and 50 mL thioglycolate flow medium,respectively.A tube containing 15 mL of sulfur glycolate fluid medium was used as control,and the growth of the bacteria in the three groups were observed.Results The five bacterial suspensions with the above two concentrations grew well in both disposable membrance filter with 50 mL of thioglycolate flow medium and test tube with 15 mL of thioglycolate flow medium.In the disposable membrane filters containing 100 mL thioglycolate flow medium,all of the bacterial suspension strains with concentration of 10-100 cfu/mL grew well,while there were 2 batches of ungrown pseudomonas aeruginosa and 3 batches of ungrown bacillus subtilis were found in the bacterial suspension strains with concentrations of 1-10 cfu/mL.Conclusion In the investigation of growth examination of thioglycolate fluid medium,it is recommended to select 100 mL disposable membrance filter and bacterial suspension strains with concentrations of 1-10 cfu/mL for reducing the probability of false negative occurrence in the sterility examination.
