妊娠高血压疾病患者血清miR-155-5p、LncRNA MALAT1表达及与糖脂代谢关系

摘　　要：目的:探讨微小RNA-155-5p(miR-155-5p)、长链非编码RNA肺癌转移相关转录本1(LncRNA MALAT1)在妊娠期高血压疾病患者血清中的表达及与糖脂代谢关系。方法:选择2019年8月-2021年12月本院收治的103例妊娠期高血压疾病患者为病例组,其中妊娠期高血压患者41例、轻度子痫前期患者35例、重度子痫前期患者27例,选择同期产前检查正常孕妇40例为对照组,qRT-PCR检测血清miR-155-5p、LncRNA MALAT1表达,检测血清糖脂代谢指标水平,Pearson法分析血清miR-155-5p与LncRNA MALAT1表达相关性,及与血糖(FBG)、糖化血红蛋白(HbA1c)、胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)水平的相关性。结果:与对照组比较,妊娠期高血压组、轻度子痫前期组、重度子痫前期组患者血清miR-155-5p、FBG、HbA1c、TG、TC、LDL-C水平升高,且随疾病程度增加而升高,LncRNA MALAT1、HDL-C水平降低,且随疾病程度增加而降低(均P<0.05);相关性分析显示,血清miR-155-5p与LncRNA MALAT1表达呈负相关性,miR-155-5p与FBG、HbA1c、TG、TC、LDL-C呈正相关,与HDL-C呈负相关;LncRNA MALAT1与FBG、HbA1c、TG、TC、LDL-C呈负相关,与HDL-C呈正相关(均P<0.01)。结论:妊娠期高血压疾病患者血清miR-155-5p高表达、LncRNA MALAT1低表达,二者具有相关性且与糖脂代谢水平和病情严重程度相关,可能作为评估患者糖脂代谢及病情变化的标志物。Objective:To investigate the expressions of the microRNA 155 5p(miR 155 5p) and the long non coding RNA lung cancer metastasis associated transcript 1(LncRNA MALAT1) in serum of pregnant women with hypertensive disorders of pregnancy,and to study their correlation with the glucose and lipid metabolism of the women.Methods:103 pregnant women with hypertensive disorders of pregnancy who were treated in hospital were selected in observation group from August 2019 to December 2021,including 41 women with hypertensive disorders of pregnancy in group A,35 women with mild preeclampsia in group B,and 27 women with severe preeclampsia in group C.40 normal pregnant women were selected in control group during the same period.QRT PCR was performed to detect the expressions of serum miR 155 5p and LncRNA MALAT1 of the women in these groups.The levels of serum glucose and lipid metabolism indexes of the women in these groups were detected.Pearson method was performed to analyze the correlation between the serum miR 155 5p expression level of the women and their LncRNA MALAT1 expression level and their levels of fasting blood glucose(FBG),glycosylated hemoglobin(HbA1c),total cholesterol(TC),triglycerides(TG),high density lipoprotein(HDL C),and low-density lipoprotein(LDL C).Results:The levels of serum miR 155p,FBG,HbA1c,TG,TC,and LDL C of the women in the control group,in group A,in group B,and in group C had increased gradually,while the levels of LncRNA MALAT1 and HDL C of the women had decreased gradually (all P<0.05).Correlation analysis showed that the serum miR 15 5p level of the women was negatively correlated with their LncRNA MALAT1level and HDL C level,and which was positively correlated with their levels of FBG,HbA1c,TG,TC,and LDL C.The LncRNA MALAT1level of the women was negatively correlated with their FBG,HbA1c,TG,TC,and LDL C levels,and which was positively correlated with their HDL C level(all P<0.01).Conclusion:The expression of serum miR 155 5p of the pregnant women with hypertensive disorders of pregnancy is
