摘  要:目的研究新型二维色谱系统建立重症医院获得性肺炎患者中替加环素含量的测定方法。方法60例重症医院获得性肺炎患者为研究对象,建立新型二维色谱系统,包括一维液相色谱柱、中间色谱柱、二维色谱柱,测定血清中替加环素质量浓度,计算方法为内标法,观察应用效果。结果替加环素为1500~2500 ng/ml时的线性关系最好(r=0.999),符合方法学验证要求;替加环素低、中、高质量浓度的绝对回收率分别94.9%、95.3%、96.2%,符合转移回收率要求(90%~110%);日间、日内精密度均<3%。结论新型二维色谱系统是一种可靠的重症医院获得性肺炎患者中替加环素含量的测定方法,操作简便,精密度高。Objective To develop a novel two-dimensional chromatographic system for the determination of tigecycline in patients with severe hospital-acquired pneumonia.Methods 60 patients with severe hospitalacquired pneumonia were selected as study subjects,and a new two-dimensional chromatographic system was established,including one-dimensional liquid chromatographic column,intermediate chromatographic column and two-dimensional chromatographic column.The internal standard method is used to determine the concentration of tigecycline in serum.The practical effect was observed.Results The linear relationship was the best(r=0.999)when tigacycline was 1500-2500 ng/ml,which met the requirements of methodology verification.The absolute recoveries were 94.9%,95.3%,96.2%for low,medium and high concentrations of tigecycline,respectively,which met the transfer recovery requirements(90%-110%),with both daytime and intra-day precision<3%.Conclusion Novel two-dimensional chromatographic system is a reliable method for the determination of tigecycline in patients with severe hospital-acquired pneumonia.It is simple and accurate.
