摘  要:目的研究敏咳方通过分泌型糖蛋白/β-连环蛋白(Wnt/β-catenin)信号通路及免疫因子动态变化对哮喘大鼠气道上皮屏障的修复作用。方法选取36只无特定病原体级健康雄性SD大鼠,随机分为正常对照组、模型组、激素治疗组、敏咳方治疗组,每组9只。正常对照组大鼠不施加任何干预因素,自由食水;模型组大鼠采用卵清蛋白氢氧化铝溶液致敏并激发,再增加烟熏、激怒、高脂饮食等形成风痰瘀型;激素治疗组大鼠在模型组基础上加激素进行治疗;敏咳方治疗组大鼠在模型组基础上加敏咳方进行治疗。采用图像分析技术测定四组大鼠支气管管壁厚度、支气管平滑肌厚度;酶联免疫吸附测定(ELISA)法检测大鼠血清卵清蛋白特异性免疫球蛋白E(OVA-sIgE)水平,实时荧光定量聚合酶链反应(qRT-PCR)法测定无翅型MMTV整合位点家族成员7B(Wnt7B)、β-catenin的表达。比较四组大鼠Wnt7B、β-catenin及支气管管壁厚度、支气管平滑肌厚度、OVA-sIgE水平。结果正常对照组Wnt7B、β-catenin分别为(1.00±0.01)、(1.00±0.01),模型组分别为(10.03±1.23)、(1.95±0.27),激素治疗组分别为(5.32±0.42)、(1.42±0.18),敏咳方治疗组分别为(4.93±1.03)、(1.33±0.21);模型组Wnt7B、β-catenin的表达均大于正常对照组,差异有统计学意义(P<0.05);激素治疗组Wnt7B、β-catenin的表达均小于模型组,差异有统计学意义(P<0.05);敏咳方治疗组Wnt7B、β-catenin的表达均小于激素治疗组,差异有统计学意义(P<0.05)。正常对照组支气管管壁厚度、支气管平滑肌厚度及OVA-sIgE水平分别为(27.44±2.92)μm^(2)/μm、(12.58±0.99)μm^(2)/μm、(32.12±3.92)ng/ml,模型组分别为(58.12±7.71)μm^(2)/μm、(25.38±1.56)μm^(2)/μm、(117.22±19.21)ng/ml,激素治疗组分别为(41.51±3.39)μm^(2)/μm、(22.27±0.91)μm^(2)/μm、(67.59±7.92)ng/ml,敏咳方治疗组分别为(37.12±3.21)μm^(2)/μm、(20.11±0.89)μm^(2)/μm、(59.51±6.39)ng/ml�Objective To study on effect of Minke decoction on airway epithelial barrier repair in asthmatic rats through Wnt/β-catenin signaling pathway and dynamic changes of immune factors.Methods A total of 36 healthy male SD rats without specific pathogens were randomly divided into normal control group,model group,hormone treatment group and Minke decoction treatment group,with 9 rats in each group.Normal control group SD rats were given free water without any intervention factors;rats in model group were sensitized and stimulated with ovalbumin-aluminum hydroxide solution,then given smoke,irritability and high-fat diet to contribute wind phlegm stasis tye;rats in hormone treatment group were treated with hormone on the basis of model group;rats in Minke decoction treatment group were treated with Minke decoction on the basis of model group.The bronchial wall thickness and bronchial smooth muscle thickness were measured by image analysis;serum ovalbumin specific immunoglobulin E(OVA-SIGE)levels were measured by enzyme linked immunosorbent assay(ELISA);the expressions of wingless-type MMTV integration site family,member 7B(Wnt7B)andβ-catenin were measured by quantitative real-time reverse transcriptase polymerase chain reaction(qRT-PCR).The levels of Wnt7B,β-catenin,bronchial tube wall thickness,bronchial smooth muscle thickness and OVA-sIgE in the four groups were compared.Results The Wnt7B andβ-catenin were(1.00±0.01)and(1.00±0.01)in normal control group,(10.03±1.23)and(1.95±0.27)in model group,(5.32±0.42)and(1.42±0.18)in hormone treatment group,and(4.93±1.03)and(1.33±0.21)in Minke decoction treatment group.The expressions of Wnt7B andβ-catenin in model group were higher than those in normal control group,and the differences were statistically significant(P<0.05).The expressions of Wnt7B andβ-catenin in hormone treatment group were lower than those in model group,and the differences were statistically significant(P<0.05).The expressions of Wnt7B andβ-catenin in Minke decoction treatment group were lower
