摘  要:目的:建立行气坦尼卡尔胶囊中东莨菪碱与莨菪碱的含量测定方法。方法:采用固相萃取-高效液相色谱(SPE-HPLC)法,色谱柱为Kromasil C_(18)(4.6 mm×250 mm,5μm),流动相为甲醇-乙腈-0.1%乙酸钠(含0.3%四氢呋喃,冰醋酸调节pH为6.40)(20∶40∶460),检测波长210 nm,流速为1.0 mL/min,柱温为35℃。结果:氢溴酸东莨菪碱和硫酸阿托品的浓度分别在3.6256~45.3200μg/mL,9.9920~124.9000μg/mL范围内线性关系良好,平均加样回收率分别为102.54%、95.20%。结论:该方法简便、快速、准确,可用于行气坦尼卡尔胶囊的质量控制。Objective To establish a HPLC method with solid-phase extractionfor the simultaneous determination of scopolamine and hyoscyamine in XingqiTannikaer capsules.Method The determination was performed on aKromasil C_(18) column(4.6 mm×250 mm,5 μm) with the mobile phase consisting of methanol-acetonitrile-0.1% sodium acetate(containing 0.3% tetrahydrofuran,glacial acetic acid adjusted pH of 6.40)(20∶40∶460) at a flow rate of 1.0 mL/min.The detection wavelength was set at 210 nm and the column temperature was 35 ℃.Result The linear relationship of the concentration range was 3.6256-45.3000 μg/mL for scopolamine hydrobromideand 9.9920-124.9000 μg/mL for atropine sulfate.The average recovery was 102.54% for scopolamine hydrobromide and 95.20%for atropine sulfate.Conclusion This method is simple,fast and accurate,which is suitable for the quality control of XingqiTannikaer capsules.
