摘 要:目的:探讨南苍术和北苍术燥湿健脾药效作用差异。方法:将70只SD雄性大鼠随机分为10只正常对照组与60只造模组,采用“外湿侵体”、“正气损耗”、“过食肥甘”等方法建立湿阻中焦证模型大鼠。造模成功后,将成模大鼠分为模型对照组、南苍术挥发油47、372 mg/kg组、北苍术挥发油19、154 mg/kg组、香砂养胃丸926 mg/kg组。正常对照组正常饲养,造模组持续施加造模条件。各组连续灌胃相应药物或生理盐水,连续7 d。末次药后测定大鼠体质量、脾脏及胸腺的脏器系数及小肠推进率等指标;观察结肠苏木精-伊红染色病理组织切片变化;以酶联免疫吸附法检测大鼠血浆中胃泌素(GAS)、胃动素(MTL)及结肠中水通道蛋白2(AQP2)的含量;利用实时荧光定量RCR(qRT-PCR)技术检测大鼠胃中促生长素(Ghrelin)、神经肽Y(Npy)和结肠组织中P物质(Sp)、血管活性相关肠肽(Vip)mRNA转录水平的变化。结果:与正常对照组大鼠相比,模型对照组大鼠体质量增长缓慢、精神萎靡、晦暗发黄(P<0.01);与模型对照组大鼠相比,南苍术挥发油372 mg/kg组大鼠一般情况有所恢复,体质量增长加快,大便形状有所好转;且大鼠小肠推进率、胸腺指数、脾脏指数明显升高(P<0.05或P<0.01);结肠组织炎性细胞减少、腺体结构改善;胃组织中Ghrelin、Npy表达显著上调(P<0.01);结肠组织中Sp mRNA表达上调,AQP2含量显著降低(P<0.01);血浆中GAS、MTL含量显著升高;南苍术挥发油47、372 mg/kg组大鼠的Vip mRNA表达均显著下调(P<0.01);北苍术挥发油154 mg/kg组大鼠血浆中MTL含量明显升高(P<0.05),北苍术挥发油19、154 mg/kg组大鼠结肠组织中Vip mRNA表达均显著下调(P<0.01)。南苍术挥发油372 mg/kg组对小肠推进率、胸腺指数、脾脏指数、Ghrelin、Npy、Sp、Vip mRNA表达的调控能力明显强于北苍术挥发油154 mg/kg组。结论:南、北苍术挥发油对湿阻中焦证均有治疗作用,�Objective:To compare the Zaoshijianpi(燥湿健脾)effect between Atractyiodes lancea and A.chinensis.Methods:Seventy SD male rats were randomly assigned into a normal control group(n=10)and a modeling group(n=60).The rat model of Shizuzhongjiao(湿阻中焦)syndrome was established with the methods of Waishiqinti(外湿侵体),Zhengqisunhao(正气损耗),and Guoshifeigan(过食肥甘).After successful modeling,the model rats were assiged into a model group,A.lancea volatile oil(47 and 372 mg/kg)groups,A.chinensis volatile oil(19 and 154 mg/kg)groups,and a Xiangsha Yangwei Pills(926 mg/kg)group.The normal group was fed in a conventional manner,and the modeling conditions were continuously applied to the modeling group.Each group was administrated with corresponding drugs or normal saline continuously for 7 days.After the last administration,the body weight,spleen index,thymus index,and small intestine propulsion rate were measured.Hematoxylin-eosin staining was employed to observe the pathological changes of colon.Enzyme-linked immunosorbent assay was employed to determine the levels of gastrin(GAS)and motilin(MTL)in the plasma and that of aquaporin 2(AQP2)in the colon.Real-time fluorescence quantitative RCR(qRT-PCR)was employed to measure the mRNA levels of Ghrelin and neuropeptide Y(Npy)in the stomach and substance P(Sp)and vasoactive intestinal peptide(Vip)in the colon tissue.Results:Compared with the normal control group,the rats in the model control group showed slow growth in body mass,listlessness,dullness,and yellowing(P<0.01).Compared with the model group,A.lancea volatile oil(372 mg/kg)alleviated the above symptoms,recovered the general conditions,accelerated the body weight growth,and improve the stool shape.The small intestine propulsion rate,thymus index,and spleen index in the A.lancea volatile oil(372 mg/kg)group were higher than those in the model group(P<0.05 or P<0.01).Furthermore,A.lancea volatile oil(372 mg/kg)reduced the inflammatory cells in the colon tissue,improved the glandular struc
