摘 要:利用基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption ionization-time of flight mass spectrometry, MALDI-TOF MS)方法构建不同畜肉的蛋白质分子指纹图谱数据库,用于鉴别猪、牛、羊源性成分。样品用磷酸盐缓冲液提取、净化后用MALDI-TOF MS法测定。采用Chem Pattern软件对不同畜肉蛋白图谱进行化学计量学分析。结果表明,通过多元统计分析,猪、牛、羊3种不同畜肉之间MALDI-TOF MS图谱具有显著差异,建立样品分类识别模型准确鉴别不同畜肉动物源性成分。建立基于蛋白质组学的猪、牛、羊源性成分鉴定系统,具有准确度高、重现性好的优点,能被广泛应用于猪、牛、羊源性成分的鉴别。Protein molecular fingerprint database of different livestock meat was constructed by matrix-assisted laser desorption ionization-time of flight mass spectrometry(MALDI-TOF MS),which was used to identify animal-derived ingredients of pork,beef and mutton.Samples were extracted with phosphate buffer,purified and then determined by MALDITOFMS.Chem Pattern software was used to analyze the protein profiles of different livestock meat by chemometrics.The results showed that through multivariate statistical analysis,there were significant differences in MALDI-TOF MS maps among pork,beef and mutton.The establishment of sample classification and identification model could accurately identify different animal-derived components.The identification system of pork,beef and mutton based on proteomics was established,which had the advantages of high accuracy and good reproducibility.It could be widely applied in the identification of animalderived components of pork,beef and mutton.
