摘 要:目的:研究亚硒酸钠对羊肚菌Y2(Morchella esculenta Y2)菌株液体发酵培养的影响,筛选羊肚菌富硒液体发酵的亚硒酸钠最适添加量。方法:在培养基中添加不同浓度的亚硒酸钠,通过测定固体培养羊肚菌菌丝生长速度、液体发酵培养菌丝生物量、胞内总蛋白含量、胞内可溶性糖含量以及硒富集率等指标,探讨不同浓度亚硒酸钠对羊肚菌液体发酵的影响。结果:高质量浓度亚硒酸钠(≥30μg/mL)对羊肚菌Y2菌株菌丝生长具有抑制作用;较低质量浓度(≤20μg/mL)则会促进生物量和富硒率增加;其质量浓度为20μg/mL时,固体培养基中的菌丝生长速度最快,液体发酵生物量为5.500 g/L、菌丝体胞内总蛋白含量为2.25 mg/g、胞内可溶性糖含量为161.6 mg/g、胞内有机硒含量达到0.180 mg/g、富硒率为10.84%。结论:羊肚菌Y2菌株液体发酵的富硒性能良好,菌丝体生物量和胞内可溶性糖含量相较于未加硒组有所提高,为富硒羊肚菌在食品和医药方面的进一步研究提供参考。Objective:To study the effect of sodium selenite on the growth and bioactive substanances of Morchella esculenta Y2 in liquid culture condition,and optimize the additive amount of sodium selenite in the selenium-enriched liquid fermentation by Morchella esculenta.Methods:By adding different concentrations of sodium selenite to the medium,the growth rate of mycelia in solid media,the biomass in liquid fermentation culture,total intracellular protein content,intracellular soluble sugar content and selenium enrichment rate were measured,to evaluate different concentrations of sodium selenite on the liquid fermentation of Morchella esculenta.Results:High concentration of sodium selenite(≥30μg/mL)inhibited the mycelial growth of Morchella esculenta Y2,while low concentration(≤20μg/mL)promoted the increase of biomass and selenium enrichment rate.When the concentration of sodium selenite in the culture medium was 20μg/mL,the maximum biomass(5.500 g/L)was obtained,with the total intracellular protein content of 2.25 mg/g,the intracellular soluble sugar content of 161.6 mg/g,the intracellular organic selenium content of 0.180 mg/g,and the selenium enrichment rate of 10.84%.Conclusion:The strain Morchella esculenta Y2 has good selenium enrichment ability under liquid fermentation condition,and the mycelial biomass and intracellular soluble sugar content is incereased compared to the control group,which may provide data reference for the use of selenium-rich Morchella esculenta in food and medicine.
