摘 要:【目的】分析枳壳不同生育期根际土壤细菌群落组成及其与土壤环境因子的关系,揭示影响土壤细菌群落结构的关键环境因子,为枳壳合理的栽培管理和土壤培育提供科学指导。【方法】分别在枳壳花期、坐果期、采收期和越冬期采集根际土壤,采用高通量测序技术测定和分析细菌的群落组成及多样性变化特征。同时,采用化学分析方法对土壤理化性质和酶活性进行测定,并运用R语言软件对细菌群落和土壤环境因子进行RDA和相关性热图分析。【结果】枳壳采收期根际土壤pH值显著高于其他生育期(P<0.05,下同);土壤全氮、有效钾含量和过氧化氢酶活性均表现为坐果期显著高于花期和采收期;高通量测序共得到29 240个细菌OTUs,分属于24门、72纲、158目、253科、409属;土壤细菌群落丰富度在枳壳生育期中呈先升后降的变化趋势,在坐果期最高;在门水平上,变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)、绿弯菌门(Chloroflexi)、放线菌门(Actinobacteria)和芽单胞菌门(Gemmatimonadetes)为优势菌门,占总丰度的82.53%~88.49%;在属水平上,鞘氨醇单胞菌属(Sphingomonas)、RB41、MND1、硝化螺菌属(Nitrospira)、Subgroup_10、Haliangium、Bryobacter和Dongia属为优势菌属,占总丰度的13.16%~19.49%;其中Dongia属菌群在坐果期明显增加,而Subgroup_10属菌群在越冬期显著增加。相关性分析表明,Haliangium属、Bryobacter属和溶杆菌属(Lysobacter)的丰度与土壤pH、有机质含量和脲酶活性呈显著正相关;而Subgroup_10属丰度与土壤有效氮含量呈显著负相关。【结论】枳壳根际土壤理化性质、酶活性及根际细菌群落组成均随生育期变化而发生波动,土壤pH和有效氮含量是影响土壤根际细菌群落结构变化的重要环境因子。【Objective】The study aimed to analyze the composition of rhizosphere soil bacterial community in different growth stages of Fructus aurantii and its relationship with soil environmental factors,reveal the key environmental factors that affect the structure of soil bacterial community,and provide scientific guidance for rational cultivation management and soil cultivation of F.aurantii.【Method】The rhizosphere soil samples of F.aurantii at flowering,fruit setting,harvesting and overwintering stage were collected respectively.Then the diversity index and community composition of bacteria was determined by high-throughput sequencing technology.The soil physicochemical properties and enzyme activities were measured by chemical analysis methods.The correlations between bacterial community structure and factors were analyzed by R software.【Result】The soil pH was significantly higher in the harvesting stage than in other stages(P0.05,the same as below);The total nitrogen,available potassium and activity of catalase were significantly higher in fruit setting stage than in flowering and harvesting stage.A total of 29 240 bacterial OTUs were obtained by high-throughput sequencing,which belonged to 409 genus,253 families,158 orders,72 classes and 24 phyla.The Chao1 index was higher in fruit setting stage than other stages.At phylum level,the dominant was Proteobacteria,Acidobacteria,Chloroflexi,Actinobacteria and Gemmatimonadetes,which accounted for 82.53%-88.49%.At the genus level,the dominant was Sphingomonas,RB41,MND1,Nitrospira,Subgroup_10,Haliangium,Bryobacter and Dongia,which accounted for 13.16%-19.49%.The abundance of Dongia was higher in fruit setting stage than other stages,while the Subgroup_10 was significantly increased in overwintering stage.The correlation analysis showed that the abundance of Haliangium,Bryobacter,Lysobacter were significantly positively correlated with pH,organic matter and activity of urease,while the Subgroup_10 was significantly negatively correlated with available nitrogen.�
