配合饲料替代冰鲜鱼对高体鰤幼鱼生长及肝脏转录组的影响

摘  要:【目的】探讨配合饲料替代冰鲜鱼条件下,高体鰤(Seriola dumerili)幼鱼生长及肝脏组织代谢相关基因的表达,揭示配合饲料影响高体生长、代谢的分子机制,为其精准配合饲料开发提供参考。【方法】分别将以冰鲜鱼与配合饲料喂养60 d的高体幼鱼肝脏组织进行转录组测序分析,筛选差异表达基因(DEGs),富集分析GO和KEGG信号通路,RT-qPCR检测不同饵料喂养后高体肝脏中生长及代谢相关基因的表达。【结果】高体肝脏转录组测序获得高质量数据(cleanreads)共247272212,各样品Q_(30)均不低于94.11%;与冰鲜鱼组对比,配合饲料组共筛选到144个DEGs,其中81个DEGs表达上调,63个DEGs表达下调;KEGG通路富集分析发现,配合饲料组相对冰鲜鱼组显著富集在甾体生物合成、不饱和脂肪酸生物合成、萜类骨架生物合成、胆固醇代谢、脂肪代谢等脂质合成与代谢通路;转录组结果显示,配合饲料组与冰鲜鱼组肝脏中生长相关基因无显著差异,而代谢相关基因表达显著上调,RT-qPCR结果与转录组结果相一致。【结论】以配合饲料替代冰鲜鱼饲喂高体幼鱼,幼鱼生长差异不显著,但肝脏的脂质代谢能力显著提升。【Objective】The aim of this study is to investigate the growth and the expression of metabolism-related genes in the liver tissue of juvenile Seriola dumerili when chilled fish were replaced by formula feed,and reveal the molecular mechanism of the effect of the formula feed on the growth and metabolism of S.dumerili,and provide reference for development of its precise formula feed.【Method】The liver tissue of juvenile S.dumerili fed with different diets for 60 days was subjected to transcriptome sequencing analysis,screening differentially expressed genes(DEGs),GO and KEGG signaling pathway enrichment analysis was performed and RT-qPCR was used to detect the expression of growth and metabolism-related genes in the liver of S.dumerili fed with different diets.【Result】A total of 247272212 clean reads were obtained from the transcriptome sequencing of the liver of S.dumerili,and the percentage of Q_(30) bases in each sample was not less than 94.11%.Compared with the chilled fish group,144 DEGs were screened in the formula feed group,of which 81 DEGs were up regulated and 63 DEGs were down-regulated.KEGG pathway enrichment analysis showed that the formula feed group was significantly enriched in lipid synthesis and metabolic pathways such as steroid biosynthesis,unsaturated fatty acid biosynthesis,terpenoid skeleton biosynthesis,cholesterol metabolism,and fat metabolism compared with the chilled fish group.The transcriptome results showed that there was no significant difference in growth-related genes between the compound feed group and the chilled fish group,while the expression of metabolism-related genes was significantly up regulated,which was consistent with the results of RT-qPCR.【Conclusion】These results indicate that replacing chilled fish with formula feed has no significant effect on the growth of juvenile S.dumerili,but it will significantly improve the lipid metabolism ability of the liver of juvenile S.dumerili.
