摘  要:[目的]明确广东省深圳市某养殖场黄喉拟水龟的突发性死亡原因。[方法]采用平板划线分离法对患病龟的肝脏、脾脏等病灶组织分离到一株优势菌,命名为GZW01,对分离菌进行形态特征和生理生化特征的鉴定,利用通用引物27F、1492R对16S rDNA序列扩增,采用DNAMAN及MEGA 6.0等软件进行生物信息学分析该病原菌分类地位。动物回归感染试验验证其致病性,药物敏感试验筛选有效的治疗防控方法。[结果]菌株GZW01为杆状革兰氏阴性菌,其氧化酶活性、葡萄糖酸盐试验、甘露醇、甘露糖、蔗糖、阿拉伯糖、肌醇、覃糖、MR、VP、精氨酸脱羧酶等生理生化鉴定为阳性,可使葡萄糖产气;与NCBI上维氏气单胞菌(Aeromonas veronii)具有99%同源性;回归感染试验表明菌株GZW01对黄喉拟水龟具有强致病性;药敏结果显示,分离菌株对青霉素G、苯唑西林、氨苄西林、四环素、万古霉素、克林霉素6种药物耐受,对克拉霉素药物中度敏感,对庆大霉素、红霉素、氧氟沙星、氨曲南等28种抗生素高度敏感。[结论]引发深圳市某养殖场黄喉拟水龟的突发性死亡的致病菌为维氏气单胞菌。[Objective]To identify the sudden death causes of Mauremys mutica in a breeding farm in Shenzhen,Guangdong Province.[Method]A dominant strain named GZWO1 was isolated from the liver,spleen and other focal tissues of the infected Mauremys mutica by the scribing plate separation method.The morphological characteristics,physiological and biochemical characteristics of the isolated strain were identified,and the general primers were used:16SrDNA sequence was amplified by 27F and 1492R,and the classification status of the pathogen was analyzed by using DNAMAN and MEGA-6.0 software.The pathogenicity of the pathogen was verified by animal regression infection test,and effective treatment and prevention methods were screened by drug sensitivity test.[Result]Strain GZW01 was a rod-like gram-negative bacterium.Its enzyme activity,gluconate test,mannitol,mannose,sucrose,arabinose,inositol,Tanose,MR,VP,arginine decarboxylase and other physiological and biochemical tests were positive,which could produce glucose gas.It had 99%homology with Aeromonas veronii on NCBI.The regression infection test showed that strain GZW01 was highly pathogenic to Tenebrio flavus.The results of drug sensitivity showed that the isolated strain was resistant to penicillin G,benzoxicillin sodium,ampicillin,tetracycline,vancomycin,clindamycin,and a total of six drugs.It was moderately sensitive to clarithromycin;and highly sensitive to 28 antibiotics such as gentamicin,erythromycin,ofloxacin and amtrannan.[Conclusion]Aeromonas veronii was the pathogenic bacterium that caused sudden death of the Mauremys mutica in a breeding farm in Shenzhen City.
