摘  要:目的基于转化生长因子(transforming growth factor,TGF)β1/Smad通路探讨舒筋健腰丸对黄韧带纤维化模型大鼠的影响和作用机制。方法40只SD大鼠随机分为正常组,假手术组,模型组,舒筋健腰丸低、高剂量组,每组8只,采用腰椎失稳法建立黄韧带纤维化大鼠模型,造模成功后各组大鼠给予相应药物灌胃,8周后对大鼠黄韧带行能谱Mcro-CT检查与Masson染色,免疫组化和荧光定量PCR检测黄韧带组织中TGF-β1、Smad2/3、Smad4、Smad7、Ⅰ型胶原蛋白(CollagenⅠ)和Ⅲ型胶原蛋白(CollagenⅢ)的蛋白及mRNA表达。结果与正常组比较,模型组大鼠黄韧带胶原蛋白断裂、扭曲、堆积,胶原容积分数(collagen volume fraction,CVF)显著升高,TGF-β1、Smad2/3、Smad4、CollagenⅠ和CollagenⅢ蛋白与mRNA表达显著增加,smad7蛋白与mRNA表达显著下降(P<0.05);与模型组比较,舒筋健腰丸高剂量组上述指标均有明显改善(P<0.05)。结论舒筋健腰丸能够有效抑制模型大鼠黄韧带纤维化进程,减少胶原堆积、扭曲和断裂,其作用可能与激活TGF-β1/Smad通路有关。Objective Based on transforming growth factor(TGF)-β1/Smad pathway to explore the effect and mechanism of Shujin Jianyao pill on ligamentum flavum fibrosis model rats.Methods 40 SD rats were randomly divided into normal group,sham operation group,model group,Shujin Jianyao low-dose group and high-dose group,with 8 rats in each group.A rat model of ligamentum flavum fibrosis was established by the destabilization method.After successful modeling,rats in each group were given corresponding drugs by gavage.After 8 weeks,the ligamentum flavum of rats in each group was detected by Mcro-CT and Masson staining.The expressions of TGF-β1,Smad2/3,Smad4,Smad7,CollagenⅠ,CollagenⅢprotein and mRNA were detected by immunohistochemistry and fluorescent quantitative PCR.Results Compared with the normal group,the collagen of the ligamentum flavum in the model group rats was fractured,twisted,and stacked.Collagen volume fraction(CVF)was significantly higher,the protein and mRNA expressions of TGF-β1,Smad2/3,Smad4,collagen I and collagenⅢwere significantly increased,while Smad7 protein and mRNA expression were significantly reduced in the model group rats(P<0.05).Compared with the model group,all the indexes above were improved significantly in rats of Shujin Jianyao pill high dosage group(P<0.05).Conclusion Shujin Jianyao pill can effectively inhibit the progression of ligamentum flavum fibrosis in model rats,and reduce collagen accumulation,distortion,and rupture.The mechanism may be related to the activation of TGF-β1/Smad pathway in the ligamentum flavum.
