隔药饼灸对慢性疲劳综合征大鼠免疫蛋白及T细胞调节因子Tim-3、LAG-3的影响

摘　　要：目的探讨隔药饼灸治疗慢性疲劳综合征(chronic fatigue syndrome,CFS)的作用机制。方法50只SD大鼠随机分为空白组、模型组、隔药饼灸组、假药饼灸组、中药灌胃组,每组各10只。采用复合应激方法造模21 d,行为学检测CFS模型,隔药饼灸组与假药饼灸组大鼠取“神阙”“关元”“足三里”“期门”穴进行相应干预,中药灌胃组大鼠给予逍遥散悬浊液灌胃,10 d后ELISA法检测各组大鼠血清中免疫球蛋白(Ig)A、IgG及补体C3含量;苏木精-伊红染色观察肝脏、脾脏组织形态学变化;实时荧光定量PCR法检测股四头肌Tim-3、LAG-3 mRNA表达水平。结果与空白组比较,模型组大鼠肝脏、脾脏病理改变明显,血清中IgA、IgG、C3含量显著降低,股四头肌Tim-3、LAG-3 mRNA水平显著升高(P<0.05)。与模型组比较,隔药饼灸组与中药灌胃组大鼠上述指标均有明显改善(P<0.05),假药饼灸组大鼠仅出现Tim-3 mRNA水平下降(P<0.05)。结论隔药饼灸可能通过提高CFS模型大鼠IgA、IgG及补体C3水平,降低免疫负调节因子Tim-3、LAG-3 mRNA水平,从而恢复机体免疫功能,增强免疫力,起到治疗CFS的作用。Objective To observe the effect of medicinal cake moxibustion treatment on rats with chronic fatigue syndrome(CFS),and to explore its mechanism.Methods 50 SD rats were randomly divided into blank group,model group,medicinal cake moxibustion group,sham medicinal cake moxibustion group and herbal gavage group,with 10 rats in each group.The rats in medicinal cake moxibustion group and the sham medicinal cake moxibustion group were given corresponding intervention respectively,which were placed in“Shenque”“Guanyuan”“Zusanli”“Qimen”.Xiaoyao powder suspension was given orally to rats in the herbal gavage group.After 10 days,the serum levels of IgA,IgG and complement C3 were detected by ELISA,the morphological changes of liver and spleen were observed by HE staining,muscle Tim-3 and LAG-3 mRNA levels were detected by Real-time quantitative PCR.Results Compared with blank group,the pathological changes of liver and spleen were obvious,the contents of IgA,IgG and C3 were significantly decreased,and the mRNA levels of Tim-3 and LAG-3 were significantly increased in rats of model group(P<0.05).Compared with model group,the above indexes in rats of medicinal cake moxibustion group and herbal gavage group were significantly improved(P<0.05),and only Tim-3 mRNA level was decreased in rats of sham medicinal cake moxibustion group(P<0.05).Conclusion Medicinal cake moxibustion may improve the levels of IgA,IgG and complement C3 in CFS model rats,and reduce the mRNA levels of immune negative regulators Tim-3 and LAG-3,thereby restore the immune function of the body,enhance immunity,and play a role in the treatment of CFS.
