摘　　要：随着全球人口逐渐老龄化,延缓衰老及防治其相关疾病成为迫切需要解决的社会问题。衰老是一种容易引发多种疾病的病理过程,衰老与这些疾病之间存在共同发生机制。因此,探索衰老及其相关疾病的发生机制和寻求高效的抗衰老药物已成为当前研究的热点。真核翻译起始因子5A(eukaryotic translation initiation factor 5A, eIF5A)是受羟腐胺赖氨酸化(hypusination)调节的一种翻译起始因子,多胺中的亚精胺(spermidine, SPD)是eIF5A发生羟腐胺赖氨酸化修饰的底物。The eukaryotic translation initiation factor 5A(eIF5A)is a ubiquitously expressed and highly con‐served protein among eukaryotes.The polyamine family member,spermidine,induces a post-translational hypusination modification in eIF5A.This process involves transferring the 4-aminobutyl segment of spermidine to the 50th lysine(Lys50)residue of the eIF5A precursor,leading to its hypusination and subsequent activation.Contemporary research in‐dicates that the activated eIF5A(hypusinated eIF5A)not only facilitates general translation initiation,elongation,and ter‐mination,but also significantly influences the aging process and related diseases by modulating autophagy,apoptosis,mi‐tochondrial functionality,and inflammatory reactions.This review delves into the polyamine-driven eIF5A-hypusination pathway's implications in age-associated ailments,such as diabetes,immune system aging and neurological dysfunctions,aiming to offer valuable insights for researchers in these domains.
