摘 要:目的应用Olink靶向蛋白质组学技术筛选高原习服和平原人血清中细胞代谢相关差异表达蛋白,了解其在机体组织或细胞缺氧适应性代谢中的功能。方法纳入2020年12月至2021年7月移居平均海拔4500 m、居住时间为6~12个月的高原习服男性40例(高原习服组),以及与之相匹配的久居平原的健康汉族男性40例(对照组)作为研究对象,收集两组基础资料和血压、心率、血氧饱和度等指标,采用Olink靶向蛋白质组学技术检测血清中92种细胞代谢相关蛋白的差异表达情况并进行富集分析。结果与平原对照组相比,高原习服组舒张压、心率较高,血氧饱和度较低,差异均有统计学意义(P<0.05);血清中细胞代谢相关的差异表达蛋白有34个,其中上调16个(P<0.05),下调18个(P<0.05);差异表达蛋白在阿米巴病、嘧啶代谢、溶酶体、细胞凋亡、胰岛素代谢、细胞黏附分子等通路显著富集,还有一些为氮素代谢、精氨酸生物合成、烟酸盐和烟酰胺代谢、磷酸戊糖途径等通路中的关键蛋白。结论高原习服人群血清中多种与细胞缺氧代谢相关的蛋白表达发生变化,表明机体组织或细胞发生缺氧适应性代谢改变。Objective To screen the differentially expressed proteins related to cell metabolism in human serum of plateau acclimatized population and plain habitants by Olink targeted proteomics technology,and to understand their function in the adaptive metabolism of tissue or cell hypoxia.Methods A total of 40 plateau acclimatized males who migrated to an average altitude of 4500 meters and lived for 6-12 months from December 2020 to July 2021(plateau acclimatization group),and 40 matched healthy Han Chinese males(plain control group)who have been living in the plain for a long time were selected as the research subjects.The basic data and blood pressure,heart rate,and blood oxygen saturation of the subjects were collected.Olink targeted proteomics technology was used to detect the differential expression of 92 cell metabolism-related proteins in serum and enrichment analysis was performed.Results Compared with the plain control group,the diastolic blood pressure and heart rate of the plateau acclimatization group were higher,and there were 34 differentially expressed proteins related to cell metabolism in the serum of the plateau acclimatization group,of which 16 were up-regulated(P<0.05)and 18 were down-regulated(P<0.05).Differentially expressed proteins were significantly enriched in pathways such as amoeba disease,pyrimidine metabolism,lysosome,cell apoptosis,insulin metabolism,and cell adhesion molecules,and some were key proteins in pathways such as nitrogen metabolism,arginine biosynthesis,nicotinate and nicotinamide metabolism,and pentose phosphate pathway.Conclusion The expression of various proteins related to hypoxia metabolism in serum of plateau acclimatized population changes,indicating that hypoxia adaptive metabolism changes occur in body tissues or cells.
