摘 要:文章基于高通量测序技术对污水处理厂微生物群落多样性进行分析研究,并针对污水处理脱氨氮效率低,产生二级污染物等问题,筛选分离出同步硝化好氧反硝化菌实现高效脱氮,探索其脱氮特性,为强化微生物法处理高氨氮废水提供新的菌源。微生物多样性分析结果显示,脱氮系统具有丰富的群落多样性,其主要菌门为变形杆菌门(Proteobacteria),主要优势菌属为陶厄氏菌属(Thauera sp.)。从污水处理系统的活性污泥中分离鉴定高效脱氮菌株。通过形态观察和16S rDNA基因序列比对,鉴定分离菌株为康德拉氏副球菌(Paracoccus kondratievae);测定不同氮源降解过程中菌体生长量变化和脱氮效率,分析其同步硝化好氧反硝化性能,结果显示,其24 h内的氨氮去除率为99.91%,将菌株应用于实际污水处理中,其总氮、氨氮、硝氮脱除率分别为60.70%,88.77%和86.35%。Based on the high-throughput sequencing technology,the microbial community diversity of sewage treatment plant was analyzed and studied.In view of the low efficiency of ammonia denitrification in sewage treatment and the generation of pollutants,simultaneous nitrification aerobic denitrification bacteria were screened and isolated to achieve efficient nitrogen removal,and their denitrification characteristics were explored to provide a new source of bacteria for the enhanced microbial treatment of high ammonia nitrogen wastewater.The results of microbial diversity analysis showed that the denitrification system had a rich community diversity,the main bacteria is Proteobacteria,and the dominant bacteria is Thauera sp..Isolation and identification of efficient denitrifier strains from activated sludge of sewage treatment system.The strain was identified as Paracoccus kondratievae by morphological observation and 16S rDNA sequence alignment.By measuring the variation of bacterial growth and denitrification efficiency in the degradation process of different nitrogen sources,the heterotrophic nitrification aerobic denitrification performance was analyzed.The results showed that the ammonium removal within 24 h was 99.91%.The removal rates of total nitrogen,ammonia nitrogen and nitrate nitrogen were 60.70%,88.77%and 86.35%,respectively.
