摘  要:本研究旨在探究西伯利亚白刺(Nitraria sibirica)中一类重要的Na+/H+逆转运蛋白编码基因NHX家族生物信息学特征及非生物胁迫下的表达模式,为深入解析盐胁迫下西伯利亚白刺NHXs基因的功能奠定基础。本研究基于西伯利亚白刺全基因组数据,以拟南芥(Arabidopsis thaliana)NHXs基因序列为参考获得西伯利亚白刺NHXs基因。在西伯利亚白刺中共鉴定出12个NHXs成员,基于亚细胞定位预测结果将NsNHXs分成3个不同的系统发育组,分别分布在5、 6、 7、 8、 12号染色体上。结果显示,NsNHXs氨基酸长度在408~1 190 aa,分子量在44.460 00~130.780 74 kDa,等电点在5.36~8.86,亲水系数在0.114~0.713,具有8~12个跨膜螺旋结构。物种内共线性分析发现6对存在共线性关系的基因,物种间共线性分析结果显示,在西伯利亚白刺和拟南芥之间共鉴定出6对直系同源基因。基因表达热图显示,NaCl处理后,NsNHX4和NsNHX7基因表达水平最高。NaCl和CaCl2混合处理下,NsNHX4和NsNHX7基因表达水平最高,且相较于单一NaCl溶液处理基因表达水平更高,NsNHX1和NsNHX7在转录水平上表达均上调。400 mmol/L NaCl胁迫处理下西伯利亚白刺叶片和根中NsNHX家族基因的实时定量PCR结果表明,NsNHX基因家族的表达存在组织差异性,NsNHX1和NsNHX7在西伯利亚白刺叶片和根中表达均上调,NsNHX4在西伯利亚白刺叶片组织中表达上调,而在根中表达下调,NsNHX1、NsNHX4和NsNHX7基因可能在植物耐盐性调控中发挥重要作用。本研究为盐胁迫下西伯利亚白刺NHX家族功能研究提供了重要信息,也为进一步研究盐胁迫调控机制提供了候选基因。The purpose of this study was to explore the bioinformatics characteristics of an important Nat/H*antiporter gene NHX fam-ily and its expression pattern under abiotic stress in Nitraria sibirica.This study will provide a basis for further analysis of the function of NHXs genes under salt stress.Based on the whole genome data of N.sibirica,the NHX genes in N.sibirica were obtained by using the Arabidopsis thaliana NHXs gene sequence as a reference,and a total of 12 NHXs members were identified in this study.NsNHXs were divided into three distinct phylogenetic groups based on subcellular localization prediction,distributed on chromosomes 5,6,7,8 and 12 respectively.The amino acid length of NsNHXs was 408~1190 aa,the molecular weight of NsNHXs was 44.46000~130.78074 kDa,and the isoelectric point was 5.36~8.86.The hydrophilic coefficient was 0.114~0.713,with 8~12 transmembrane helical structures.By collinearity analysis within species,6 pairs of genes were found to have collinearity relationships.The results of interspecies collinearity analysis showed that a total of 6 pairs of orthologous genes were identified between Nitraria sibirica and Arabidopsis thaliana.The gene expression heatmap showed that NsNHX4 and NsNHX7 gene expression levels were the highest after NaCl treatment.Under mixed NaCl and CaCl,treatment,NsNHX4 and NsNHX7 gene expression levels were the highest,and compared with single NaCl solution treatment,NsNHXI and NsNHX7 gene expression was up-regulated at the transcriptional level.The real time quantitative PCR results of NsNHX family genes in the leaves and roots of N.sibirica treated with 400 mmol/L NaCI showed that there were tissue differences in the expression of NsNHX family genes,and the expression of NsNHXI and NsNHX7 was up-regulated in the leaves and roots of N.sibirica.The expression of NsNHX4 was up-regulated in the leaf tissues and down-regulated in the roots.This suggests that the NsNHXI,NsNHX4 and NsNHX7 genes may play an important role in the regulation of plant salt tolerance.This st
