摘 要:目的 挖掘痤疮丙酸杆菌外排泵耐药相关基因及通路。方法 对阿奇霉素外排表型阳性的耐药痤疮丙酸杆菌,使用外排泵抑制剂羰基氰氯苯腙(CCCP)处理,分别提取处理前后的痤疮丙酸杆菌总RNA进行转录组测序与基因本体(GO)、基因组百科全书(KEGG)分析。结果 转录组测序中共发现755条差异表达基因。其中下调表达427条(56.56%),在GO数据库中注释到250个,在KEGG数据库中注释到182个,上调表达的基因有328条(43.44%),在GO数据库中注释到266个,在KEGG数据库中注释到236个。KEGG分析中下调表达基因富集最显著的通路为ABC转运蛋白通路,此通路共55个基因,差异表达基因含有27个,其中下调的前十个基因为:gtsC、ngcF、cycB、cebF、ngcE、gtsB、gtsA、oppB、oppA、ecfT。结论 CCCP对阿奇霉素外排阳性耐药痤疮丙酸杆菌的影响,主要以下调基因表达为主,且下调表达基因中以ABC转运蛋白通路最为显著,推测痤疮丙酸杆菌外排泵耐药与该通路及相关基因有密切关系。Objective To explore the genes and pathway related to drug resistance of Cutibacterium acnes efflux pump.Methods The drug-resistant Cutibacterium acnes positive for azithromycin efflux pump phenotype was treated with efflux pump inhibitor CCCP.Total RNA of Cutibacterium acnes before and after treatment was extracted for transcriptomic sequencing and Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) analysis.Results A total of 755 differentially expressed genes(DEGs) were identified by transcriptome sequencing.Among them,427 genes(56.56%) were down-regulated,250 were annotated in the GO database,182 were annotated in the KEGG database;328 genes(43.44%) were up-regulated,266 were annotated in the GO database,and 236 were annotated in the KEGG database.In KEGG analysis,the most significantly down-regulated gene enrichment pathway was ABC transporter pathway,which contained 55 genes with 27 DEGs,among which the top 10 down-regulated genes were gtsC,ngcF,cycB,cebF,ngcE,gtsB,gtsA,oppB,oppA,and ecfT.Conclusion The effect of CCCP on drug-resistant Cutibacterium acnes positive for azithromycin efflux pump phenotype is mainly manifested as down-regulated gene expression,and the ABC transporter pathway is the most significant down-regulated gene.It is speculated that drug resistance of Cutibacterium acnes efflux pump is closely related to this pathway and its related genes.
