摘 要:目的探讨补阳还五汤联合骨髓间质干细胞(MSCs)移植治疗脊髓损伤的作用机制。方法选择8~10周的健康清洁级成年Sprague-Dawley大鼠45只,随机分为假手术组,空白对照组、补阳还五汤组、MSCs移植组、补阳还五汤+MSCs组,每组9只。采用改良式的Allen’s打击法造模,分别于术后1、3、5周后通过改良Tarlov评分和斜板试验判断其对神经功能的影响,并且免疫组化检测脊髓组织中Brdu的水平,Western检测p-JAK2、p-STAT3、GP130、IL-6的蛋白表达水平。结果补阳还五汤和MSCs移植能够改善脊髓损伤大鼠的脊髓损伤程度,提高了脊髓损伤大鼠的Tarlov评分,倾斜平面临界角度也明显增加,与假手术组相比,模型组中p-JAK2、p-STAT3、p-AKT、GP130、IL-6的表达水平均显著上调;与模型组相比,补阳还五汤、MSCs细胞移植、补阳还五汤+MSCs组中的p-JAK2、p-STAT3、GP130、IL-6的蛋白表达水平均显著下调。脊髓损伤模型大鼠中Brdu含量与假手术组相比随着时间的增加明显降低,而补阳还五汤、MSCs细胞移植组中Brdu含量与模型组相比在不同的作用时间明显上升,MSCs细胞移植、补阳还五汤+MSCs组Brdu含量在第1周时表达最多。结论补阳还五汤和MSCs移植能够改善脊髓损伤,且能够抑制p-JAK2、p-STAT3、GP130、IL-6的蛋白表达,促进SCI后的神经再生修复功能,可能与补阳还五汤和MSCs移植能够抑制IL-6蛋白的表达,从而抑制JAK2/STAT3信号通路的表达有关。Objective To investigate the therapeutic effect of Buyang Huanwu Decoction Combined with MSCs transplantation on spinal cord injury through JAK2/STAT3 signal pathway.Methods Select 45 adult Sprague-Dawley rats of 8 to 10W healthy clean grade,It was randomly divided into sham surgery group,blank control group,yang and five soup group,MSCs transplant group,and Buyang Huanwu Decoctien+MSCs group,9 per group,using the modified Allen's strike method to mold,respectively,after 1,3,5w after the operation,the effect on nerve function was judged by improved Tarlov score and oblique plate test,and the level of Brdu in the spinal cord tissue was detected by immunohistochemistry,and the level of Brdu in the spinal cord tissue was detected in Western,p-JAK2,p-STAT3,GP130,and IL-6 The level of protein expression.Results Buyang Huanwu Decoction and MSCs transplantation can improve the degree of spinal cord injury in rats with spinal cord injury,improve the Tarlov score of rats with spinal cord injury,and significantly increase the critical angle of inclined plane.Compared with the sham operation group,the expression levels of p-jak2,p-STAT3,p-Akt,gp130 and IL-6 in the model group were significantly up-regulated;Compared with the model group,the protein expression levels of p-jak2,p-STAT3,gp130 and IL-6 in Buyang Huanwu Decoction,MSCs cell transplantation and Buyang Huanwu Decoction+MSCs groups were significantly down regulated.Compared with the sham operation group,the content of BrdU in spinal cord injury model rats decreased significantly with the increase of time,while the content of BrdU in Buyang Huanwu Decoction and MSCs cell transplantation group increased significantly at different action times compared with the model group.The content of BrdU in MSCs cell transplantation,Buyang Huanwu Decoction+MSCs group was most expressed at the first week.Conclusion Buyang Huanwu Decoction and MSCs transplantation can improve spinal cord injury,inhibit the protein expression of p-jak2,p-STAT3,gp130 and IL-6,and promote the function
