摘  要:目的探究仙灵脾颗粒不同活性部位体外抗骨质疏松作用,并开展其抗骨质疏松谱效相关性研究,揭示仙灵脾颗粒抗骨质疏松的药效成分群。方法采用HPLC建立仙灵脾颗粒不同活性部位指纹图谱,以小鼠MC3T3-E1成骨细胞为体外模型,以成骨细胞增殖率及碱性磷酸酶(ALP)活性为药效指标,评价仙灵脾颗粒体外抗骨质疏松药效;采用灰色关联度结合偏最小二乘法,建立活性部位共有峰与抗骨质疏松活性的谱效关系,探索贡献较大的成分。结果仙灵脾颗粒不同活性部位指纹图谱中共确定24个共有峰,各活性部位均具有促进成骨细胞增殖及ALP活性的作用。促进骨形成的药效成分群为19、16(宝藿苷II)、12(淫羊藿苷)、21、20(淫羊藿次苷I)、23(宝藿苷I)、22及17号峰;促进骨分化的药效成分群为12(淫羊藿苷)、19、21、22、24及14号峰。结论该研究明确了仙灵脾颗粒抗骨质疏松作用,筛选出促进骨形成及骨分化药效成分群,为建立以药效成分为指标的仙灵脾颗粒质量评价体系提供实验依据。Objective To explore the anti osteoporosis effect of different active parts of Xianlingpi granule in vitro,and to study the relationship of spectrum and anti osteoporosis activity,and reveal the pharmacodynamic component group of Xianlingpi Granule.Methods The fingerprints of different active parts of Xianlingpi granule were established by HPLC.The mouse MC3T3-E1 osteoblasts were used as an in vitro model.The osteoblast proliferation rate and alkaline phosphatase(ALP)activity were used as pharmacodynamic indexes to evaluate the anti osteoporosis effect of Xianlingpi granule.The grey correlation degree method combined with partial least square method were used to establish the spectrum effect relationship between the common peaks of active parts and the anti osteoporosis activity,and to explore the components with greater contribution.Results A total of 24 common peaks were picked up in the fingerprints of different active parts of Xianlingpi granule.The results showed that the active parts can promote osteoblast proliferation and ALP activity.The pharmacodynamic component groups that promote bone formation were 19,16(pogoside II),12(icariin),21,20(icariin I),23(pogoside I),22 and 17 peaks.The pharmacodynamic component groups that promote bone differentiation were 12(icariin),19,21,22,24 and 14 peaks.Conclusion This study confirmed the anti osteoporosis effect of Xianlingpi granule,screened out the pharmacodynamic component groups that promote bone formation and bone differentiation,and provided experimental basis for establishing the quality evaluation system of Xianlingpi granule.
